GDR ADN 2014 Chromatin folding in estrogen regulated transcription Silvia Kocanova, Mathieu Dalvai and Kerstin Bystricky Laboratoire de Biologie Moléculaire Eucaryote (LBME) – CNRS, University of Toulouse, France Variations in the three-dimensional organization of chromosomes guide genome function from gene expression to DNA repair and recombination. DNA-bound transcription factors recruit many chromatin remodeling and modifying complexes to activate transcription. How the local chromatin environment prepares for the transcription factor to associate with target sequences in response to stimuli is poorly characterized. We demonstrate that reptin / pontin and the histone variant H2A.Z are required to remodel the chromatin environment at number of estrogen responsive genes in mammary tumour cells. We further mapped chromatin folding over several hundreds of kb around estrogen responsive genes using FISH and confronted it with 5C data to establish a model of hormone-induced chromatin remodeling. Our findings provide new insight into the priming of chromatin required for transcription factor access to their target sequence.
