Experimental summary Norwich DMSO treated versus Control Yeast Randy Bielski Hans Hulsebos Joe Smith Melissa Latshaw • My hypothesis is that the genes: ICL1 , FLR1 will be up regulated or down regulated in the DMSO sample versus control • I base this hypothesis on the following information: previous research that induced environmental stress on the Saccromycies cerevisiae • This information was obtained from the following sources www.yeastgenome.org Gene ontology • My gene ICL1 has been assigned the following GO categories isocitrate lyase activity (IMP, ISS), glyoxylate cycle (TAS), cellular component unknown (ND) • My gene FLR1 has been assigned the following GO categories response to toxins, multi- drug transporter activity Detection call summary • Gene ICL1 had the following detection calls in the experiment : Was present in all eight experiments and had < .005 p value • Gene FLR1had the following detection calls in the experiment : Was present in all eight experiments and had no significant p value Fold change and p-values • Gene ICL1 had the following F.C and p-value in DMSO treated versus control Fold Change = 1.69; P-Value < .005 Gene FLR1 had fold changes of 1.15; P-value was not significant. Making Inferences • The following common interacting pathways exhibited differential gene expression: serine isocitrate lysase pathway. Making Inferences • Evidence for this pathway’s disregulation is further reinforced by the following data mined from public data bases • Please refer to next slide… thank you S. cerevisiae Pathway: serine-isocitrate lyase pathway Experimental conclusion • • The gene ICL1 I anticipated to be upregulated with the treatment of DMSO were confirmed by the data analysis and reinforced by additional data in the public realm leading me to conclude that ICL1 was significantly upregulated in DMSO yeast when compared to control. The gene FLR1 was anticipated to be disregulated with the treatment of DMSO were not confirmed by our experimental data – 1. Three reasons that could have lead to the non-confirmed result There was not a high fold change 2. Gene might have bound with Schizosaccharomyces pombe and was eliminated I am now an official UVM microarray data analyst and the results of my analysis are posted here
