5 min Insect DNA/RNA Preservation and Extraction Kit
Biofactories’ 5 min Insect DNA/RNA Preservation and Extraction Kit provides the fastest
method for the storage/preservation and isolation/purification of total DNA/RNA from insect
samples. The kit is specially designed for preservation and extraction of cellular and viral
DNA/RNA from insect such as honey bee and mosquito.
Insect DNA and RNA is an important source for species’ classification, gene expression
studies, and monitoring the presence of some pathogens. For classification, the total genome
DNA can be extracted and used for PCR assay in the presence of a species’ specific primers.
Total RNA can be used for studies of gene expression after removal of genomic DNA by DNase
treatment. Presence of a specific viral or bacterial pathogens can be determined in the presence
of the pathogen specific primers using qPCR or qRT-PCR. Extraction of both DNA/RNA from
the single sample is especially important when the sample is limited or to minimize the laborsome extraction procedures of DNA or RNA only preparation. The kit for total DNA/RNA
extraction from insect is essential for studies of arbovirus in mosquito or viral or microorganism
pathogenic studies in honeybee.
A reliable method is required not only for extraction of Nucleic acid but also preservation
and storage from insect samples. In typical case, the samples collected from field should be
stored in freezing especially for the liable RNA. Unless the sample is instantly frozen, the
quality of RNA would be not guaranteed because of the high contents of Nuclease activity in
the samples.
Insect sample can be mixed with the specially designed solution in the kit and kept on ice

for more than 2 days without any sign of DNA/RNA degradation. Furthermore, mixing with
the solution inactivates all pathogenic microorganisms of bacteria and virus. The stored
samples can be used for total DNA/RNA extraction in less than 5 minutes by 5 min Insect
DNA/RNA Preservation and Extraction Kit. The new chemistry in extraction solution would
allow purification of intact cellular and microbial DNA/RNA. The purified DNA/RNA is of
the highest integrity, and can be used in a number of downstream applications without any sign
of inhibitors. The kit can be served for an idealistic tool for classification, expression studies,
and pathogen studies of insects.


Before Starting
1. Add 42 ml of 96-100% ethanol to Insect DNA/RNA Washing Solution. Mix
well. Mark on the labels that ethanol is added. Store it at room temperature.
2. Warp the distal end of microcentrifuge tube pestle with folded paper towel and
type it. With this set up, you can apply more pressure to the pestle for more
effective tissue extraction.
3. The method for the squeezing samples by the microcentrifuge tube pestle is
linked to a video file in YouTube (coming up soon).
4. The microcentrifuge tube pestle can be reused after wash in running tap water,
spray with 70% ethanol, and wipe out with a clean paper towel
Procedures for Insect preservation
1. Transfer less than 0.2 g of insect samples to 1.5 ml microtube (not supplied). It
can be less than 5 mosquitoes or 1-2 honeybees.
2. Add 0.5 ml of Insect DNA/RNA PE Solution and store on ice (for less than 2
days) or -20oC (for more than a month) until DNA/RNA extraction.

Procedures for 5 min insect DNA/RNA extraction after preservation
Important notice;
I.
All steps in ambient temperature.
II. All centrifugation is in 13,500 rpm at room temperature.
1. Squeeze all solid mass with microcentrifuge tube pestle several times. Squeeze
slowly enough not to splash out of the solution. It is important grind the sample
completely for higher yield of Nucleic Acid.
2. Spin for 15 sec. to separate a solid pellet.
3. Transfer 500 ul of lysate to new microcentrifuge tube (Note; Carry over some
debris in this stage is not critical for following procedure).
4. Add 375 ul of isopropanol and mix well.
5. Transfer 0.7 ml of lysate to the supplied column and centrifuge for 15 sec.
6. Discard the flowthrough. Reassemble the spin column with its collection tube.
7. Repeat steps 5 to 6.
8. Apply 700 ul of DNA/RNA Washing Solution and centrifuge for 15 sec.
9. Apply 400 ul of DNA/RNA Washing Solution and centrifuge for 30 sec.
10. Replace the collection tube with a clean microcentrifuge tube (not supplied).
11. Add 50-100 ul of Elution Buffer to column.
12. Close the cap and vortex for 15 sec.
13. Centrifuge for 1 min.

Example of test
Total DNA/RNA preservation and extraction from insect using 5 min Insect DNA/RNA
Preservation and Extraction Kit. Insect samples was kept in Insect DNA/RNA PE Solution for 2
days at 4oC (lanes 1, and 2,; four Culex pipiens and lanes 3 and 4; one Apis mellifera) and used for
DNA/RNA extraction. Nucleic acid was eluted to final 100 ul of Elution Buffer and analyzed on
1.5 % agarose gel by 10 ul each.
