Biotechnology Labs Makeup Assignment Purpose: The purpose of this assignment is as a makeup for the Onion DNA, Dye/Indicator, Crime Scene DNA or Bacterial Transformation Labs. If you missed any of these labs, you may have been given this assignment as a makeup. Procedure: 1) Visit the appropriate website(s) below. Read the background information and engage in the various activities available, which are similar to the labs we did in class. What reagents (i.e. chemicals) are needed and what is the function of each reagent? (1 page) Dye/Indicator Lab Only: -how does electrophoresis work? On what basis does it separate mixtures of molecules? What kinds of things could you use electrophoresis to do? (1 page) DNA Crime Scene Lab Only: -what are RFLP’s? How do they relate to doing DNA “Fingerprinting”? What is a restriction digest? How does it relate to the result you see when you perform gel electrophoresis on your digested DNA? (1 page) Bacterial Transformation Lab Only: -what are plasmids? What is conjugation? How do scientists take advantage of bacteria’s natural processes to insert genes into the bacteria? (1 page) ALL LABS: -answer all the questions for each lab (1 page ea.) (i.e. your report should be 1 page in length for each lab!) 3) Clearly label each report with the title(s) of the Lab(s) you are making up, and submit the report to your instructor.
