Matrigel™ Basement Membrane Matrix Coated Cellware

BD BioCoat Cellware provides researchers with the ability to control in vitro cellular environments for cell
growth and differentiation under physiologically relevant conditions. Extracellular Matrix (ECM) is secreted
by cells to form interstitial matrix and basement membrane which constitutes the framework to which cells
are anchored. Basement membrane separates cells from mesenchymal connective tissue and provides the
spatial orientation and stability required for the organization and development of the characteristic histology
of specific tissues. In addition to its structural function, ECM has been recognized for the dynamic role it
plays in the regulation of cell growth, differentiation and biochemical function. ECMs also appear to function
in the sequestration, storage and presentation of growth factors. BD BioCoat ECM Coated Cellware is
suitable for serum-free or serum-containing cultures to promote cell attachment, spreading, growth and/or
differentiation of a variety of normal and neoplastic cells.
BD BioCoat Matrigel tissue culture plates contain a solution of Matrigel Basement Membrane Matrix which
is stored frozen. Matrigel Basement Membrane Matrix is a solubilized tissue basement membrane
preparation containing laminin, collagen type IV, heparan sulfate proteoglycan and entactin. At elevated
temperatures this matrix forms a "basement membrane" gel which promotes the attachment and
differentiation of normal and transformed cells. Matrigel Basement Membrane Matrix is formulated in
Dulbecco's Modified Eagle's Medium containing gentamicin and is compatible with all culture media.
Applications
BD BioCoat Cellware may be used for studies of cellular differentiation, cell matrix interactions, receptorligand binding, gene expression and regulation, embryogenesis and organ development, drug screening and
sensitivity assays and the culture of primary tumors.

MATRIX:
Matrigel Matrix is useful as a substrate for growth and differentiation of
many normal and transformed cell types, such as epithelial, endothelial,
muscle and neuronal cells. Specific applications for Matrigel Matrixtreated cellware include cell culture of parotid acinar cells1 and myogenic
cells,2 maintenance of normal morphology of sertoli cells,3 studies of
multiple drug resistance in hepatocytes4 and effects on cell function and
morphology of lung alveolar cells.5

HANDLING
AND USE:
KEEP FROZEN UNTIL USE!
Carefully remove the BD BioCoat plate from the foil bag and place on a
level surface at 2-8°C overnight. DO NOT ATTEMPT TO THAW THE
PLATE IN A WARM WATERBATH.
If the plate is not thawed on a level surface, the distribution of the
Matrigel™ Basement Membrane Matrix will not be even. This can be
easily rectified by gently rocking the plate to redistribute the coating.
To form a stable gel, place the thawed plate at 35° - 37°C for 30 minutes.
The plate may not be refrozen.
Cells grown on Matrigel Basement Membrane Matrix may be recovered
from the gel by the use of Dispase (Cat. # 354432)
QUALITY CONTROL:
BD BioCoat plates has been tested and found negative for the presence
of bacteria and fungi.
The biological activity of Matrigel Basement Membrane Matrix coated
plates has been determined in a chick dorsal root ganglia neurite
outgrowth assay. A maximal positive response was demonstrated after 48
hours in the absence of nerve growth factor.
STABILITY:
Stable for a minimum of 3 months from day of shipment when stored at
-20°C. KEEP FROZEN UNTIL USE.
