onion, pepper, potato etc.)

Observing Plant Cells
A. Onion Cell (Allium cepa L.)
1. Prepare a wet mount slide of the thin epidermis of an onion scale following the instructions below:
1. remove the outer scale from an onion bulb
2. snap backwards the onion scale between the fingers
3. strip-off the thin epidermis with the use of forceps
4. place a small piece of epidermis in a drop of water on the slide. Flatten the epidermis on
the slide using a needle. Cover with cover slip. Examine the onion wet mount slide under
the low power and high power objectives of your microscope. Look for a group of cells.
Although the cells appear to be rectangular, remember that they are three-dimensional,
that is they have length,width and thickness. Observe for the structure of the onion cells.
B. Red Pepper Cell (Capsicum frutescens)
1. Using a sharp razor blade, make a very thin section of the epidermis of a red sweet pepper. Mount
this in water and examine under LPO. Note the arrangement of the highly irregularly shaped cells
with many slides.
Observing Animal Cells
A. Cheek Cells
1. Obtain a toothpick, iodine solution, slide and cover slip.
2. Gently scraps the inner lining of your cheek with the blunt end of a toothpick. There seems to be
nothing on the toothpick but there are plenty of tiny cells attached to it.
3. Spread this on a glass slide, put a drop of iodine or methylene blue and leave it for one minute.
4. Remove excess stain by washing the slide in water. When washing, do not wipe the slide so as not
to remove the cells
5. Remove excess water by blotting the slide with a tissue paper.
6. Place the cover slip and focus under the LPO/HPO of the microscope.
