Post-translational modifications (PTMs) enable the cell to change the properties and activities of proteins after expression. Most of the PTMs are catalyzed by specialized enzymes. Practically all proteinogenic amino acids can be modified, thus PTMs considerably increase the diversity of protein structures. PTMs generate or eliminate charged moieties and thus changes the pI of a protein, e.g. by Ophosphorylation or Nε-acetylation of Lys. N-Acetylation and N-methylation of histones are tools for regulating DNA transcription, they can be reverted. Non-enzymatic PTMs can be an indication of “aging” of proteins or pathological processes.
