Specimen Preparation Guide
DNA and/or RNA
for Cancer Services
A key consideration for Personalis Cancer Analysis is to submit specimens that contain the amount of DNA and/or RNA necessary for sequencing
and analysis. The following information is being included as a guideline.
DNA
Sample Information
All DNA samples should meet the following requirements:
Personalis will supply a sample submission spreadsheet, which must be
emailed to Personalis prior to the start of the project. Please do not use
special characters in sample identification names.
• Must be extracted from human tissue samples.
• DNA must not be PCR amplified (no WGA samples).
• Must be pure, free of contaminants such as proteins, salt and organic
solvents.
Shipping Instructions
• Must be free of contaminating nucleic acids from other individuals or
other species.
• DNA samples can be shipped on either dry ice or blue ice.
• Should be quantified using a double stranded DNA specific method
such as PicoGreen®. We consider OD quantification (such as
NanoDrop™) to be inaccurate for our requirements.
• RNA samples must be shipped on dry ice.
DNA from Fresh Tissue
• Should be at a concentration of 10 ng/µl or greater
• Volume should be ≥ 50 µl
• If using microcentrifuge tubes, screw-top lids are preferred. Please
use Parafilm® to cover lid for added seal precaution and clearly
label tubes with water-resistant tags to ensure ink is not removed.
Place individual tubes into a 50 mL conical tube filled with packing
material (such as paper towels) to absorb the stress of shipping.
Fill the void with paper to avoid physical damage during transport.
DNA from FFPE Samples
• All shipments must comply with all applicable local, state, and
federal laws governing packing, marking and labeling. Blood,
fluids, and other specimens containing or suspected to contain
infectious substances must be shipped according to applicable
government and International Air Transport Association (IATA)
regulations.
• Should be at a concentration of 20 ng/µl or greater
• Ship samples using overnight shipment via either FedEx or UPS.
• Amount of DNA required is ≥ 250 ng, 500 ng preferred
• Buffer can be 1 x TE, 10mM Tris ph8 or nuclease-free water
• OD 260/280 must be between 1.8 and 2.0
• Volume should be ≥ 50 µl
• Total amount of DNA required is ≥ 500 ng, 1 µg preferred
• Buffer can be 1 x TE, 10mM Tris ph8 or nuclease-free water
• OD 260/280 must be between 1.8 and 2.0
RNA
RNA samples should meet the following requirements:
• Must be extracted from human tissue samples
• Must be at a concentration of 50 ng/µl or greater
• Volume must be a minimum of 10 µl
• Total amount of RNA required is ≥ 500 ng
• Must be in nuclease-free water
• OD 260/280 must be between 1.6 and 1.8
• Customers from U.S. should ship samples on 5 LBS OF DRY ICE
for overnight delivery from Monday through Thursday.
DO NOT SHIP ON FRIDAY.
• International customers MUST SHIP SAMPLES ON MONDAYS
ONLY WITH 20–25 LB DRY ICE.
• Ship the samples and a hard-copy of the manifest to the following
address:
