Introduction
• Hepatic fibrosis is one of the common disease in
Koreans.
• Preventing initial liver fibrogenesis may
contribute to hepatic fibrosis therapies.
• It is important to discover the mechanisms
related to the proliferation and apoptosis of
hepatic stellate cell line which has the critical
role in liver fibrogenesis.
AIM
• To compare gene expression patterns time
dependently between normal, activated and
apoptotic human hepatic stellate cell line
• To identify critical genes involved in hepatic
fibrosis
Materials & Method
• hTERT cell line : immortalized activated human
hepatic stellate cell line
• PDGF-BB : Platelet Derived Growth Factor-BB ;
a potent growth factor
• Curcumin : a phytochemical compound known to
induce apoptosis
• PDGF-BB and curcumin treatment on hTERT
cell line
• mRNA extraction of proliferation and apoptosis
induced hTERT cell line
• Microarray hybridization
• Data acquisition
• Spot preprocessing, quality inspection,
normalization, clustering & classification,
differentially expressed gene finding & biological
validation
• Gene expression confirmation by RT-PCR
