Figure S1: SDS Page gel containing cell envelope proteome fractions (15μg protein/lane) of L. plantarum WCFS1 (lane 2) and
its srtA deficient derivative (NZ7114, Lane 3). Lane 1 indicates protein molecular weight marker. For isolation of bacterial cell
envelopes, 100 mL bacterial cultures (of L. plantarum WCFS1 and NZ7114 were grown overnight (OD600 approximately 4.5) to
stationary phase of growth in 2  CDM without antibiotics. Cells were harvested by centrifugation (6000 × g, 20 min, 4 oC) and
subsequently washed once with PBS (pH 7.5). For isolation of released proteins, culture supernatants were filtered through a
cellulose-acetate filter (0.2 µm pore size, 25 mm diameter; Sigma-Aldrich, Missouri, USA), and proteins were precipitated by
the addition of trichloroacetic acid (TCA) to a final concentration of 10 %. After 2 hours incubation on ice, samples were
centrifuged (12000 × g, 45 minutes, 4 oC). Protein pellets were washed 3 times with acetone, air-dried, resuspended in water
and stored at -20 oC prior to analysis.
