CXCL9 is an inflammatory chemokine initially identified by differential screening of a cDNA library from limphokine-activated
macrophages. CXCL9 is a CXC chemokine and member of the nonELR (lacking a Glu-Leu-Arg motif in the N-terminal region) CXC
chemokine family. CXCL9 shares the CXCR3 receptor with CXCL10
and CXCL11, and these ligands differ in their receptor-binding and
activating properties; CXCL11 is more potent than CXCL10, and
CXCL10 is more potent than CXCL9.

FGF-basic (FGFb) is a member of the fibroblast growth factor (FGF)
family which includes 23 members. It is also known as fibroblast
growth factor 2, Fgf-2, Fgfb, and bFGF. FGFb is expressed in almost
all tissues and plays an important role in a variety of normal and
pathological processes, including development, wound healing,
and neoplastic transformation. FGFb is mitogenic for many cell
types, both epithelial and mesenchymal. It shows potent angiogenic activity and has been implicated in tumor angiogenesis.
FGFb also significantly promoted the proliferation of adipose-derived mesenchymal cells (AMC) and enhanced chondrogenesis in
three-dimensional micromass culture. FGFb binds to a family of
four distinct, high affinity tyrosine kinase receptors, designated
FGFR-1 to -4. In addition, FGFb binds to the extracellular matrix
and heparan sulfate (HS), which is an essential and dynamic regulator of fibroblast growth factor (FGF) signaling. Two fundamentally different crystallographic models have been proposed to
explain, at the molecular level, how HS/heparin enables FGF and
FGF receptor (FGFR) to assemble into a functional dimer on the
cell surface, although there is controversy regarding the exact
manner by which this occurs.

Interleukin-5 (IL-5) is a homodimeric, disulphide-linked protein
produced by T-cells. The native protein can have variable molecular weight as a result of glycosylation. Monomeric mouse IL-5
is a 113 amino acid protein with a reported molecular weight of
35 to 37 kD for the homodimeric protein. Mouse and human IL-5
are approximately 70% identical. IL-5 has been shown to promote
the growth of immature hematopoietic BFU-E progenitors and
stimulates the activation and differentiation of eosinophils, and
promotes the generation of cytotoxic lymphocytes. Mouse IL-5

In adult mice, IL-7/IL-7R signaling up-regulates expression of early
B cell factor (EBF), which in turn, regulates expression of B cell-specific genes required for the transition from lymphoid progenitor
to pro-B cells. IL-7 also plays a role in the development of secondary lymphoid tissues. IL-7 is necessary to specify CD8 lineage differentiation during CD4/CD8 cell fate choice in the thymus by inducing expression of the transcription factor Runx3. IL-7 induces
anti-apoptotic factors Bcl2 and Bcl-xL and inhibits pro-apoptotic
factors such as Bad and Bax. In this fashion, IL-7 induces cell activation, survival, and proliferation of T lymphocytes. In addition,
IL-7 controls T-cell size and metabolism through the activation of
PI3 kinase-dependent pathways and regulation of glucose metabolism. IL-7 also controls T cell–dendritic cell interactions that
are essential for both T-cell homeostasis and activation in vivo.
CD4 T cell lymphopenia increases the expression of circulating
IL-7, and TGF-β induces IL-7 downregulation.

Interleukin-10 (IL-10) was originally described as Cytokine Synthesis Inhibitory Factor (CSIF) by virtue of its ability to inhibit cytokine
production by Th1 clones. IL-10 shares over 80% sequence homology with the Epstein-Barr virus protein BCRFI. The biological activities of IL-10 include inhibition of macrophage-mediated cytokine synthesis, suppression of the delayed type hypersensitivity

Interleukin-12 [IL-12 (p70)] is a disulfide-linked heterodimer composed of unrelated p40 (glycosylated) and p35 subunits. IL-12 acts
as a growth factor for activated human T and NK cells, enhances
the lytic activity of human NK cells, and stimulates the production
of IFN-γ by resting human PBMC. IL-12R is formed by two chains,

factor expressed in bone marrow stromal cells. IL-7 is essential
for normal murine B cell development, and plays a key role in
regulating the homeostasis and function of T-cells. The binding
of IL-7 to its receptor induces dimerization of IL-7Ra and the common gamma chain (gc), which leads to activation of receptorassociated tyrosine Janus kinases JAK1 (IL-7R) and JAK3 (γc). The
activated JAK proteins, in turn, phosphorylate specific residues

IL-12 and IL-23 share the p40 subunit, which heterodimerizes
with IL-12 p35 or IL-23 p19 subunits, respectively, to form IL-12 or
IL-23. IL-12 p40 exists as a monomer and as a homodimer (IL-12
p80). IL-12 induction is relevant in asthmatic airway inflammation and IL-12 expression can be induced by mouse parainfluenza
type I (Sendai) virus in airway epithelial cells. In that experimental model, IL-12 induction is followed by excessive expression of
IL-12 p40 that could be further enhanced in IL-12 p35-deficient
mice. Overexpression of IL-12 p80 causes macrophage accumulation and contributes to airway inflammation and consequent
morbidity during viral bronchitis. Amplified epithelial IL-12 p40
expression and augmented concentrations of BAL fluid IL-12 p40
(but not IL-12 p70) has been detected in asthmatic subjects. Also,
it has been demonstrated that p80, but not IL-12 or p40, induces
macrophage chemotaxis that is independent of IL-12 and mediated through the cytoplasmic tail of IL-12b1. Additional studies
with transgenic mice suggest that overexpression of IL-12 p80
prior to a viral infection increases the number of resident airway
macrophages, which primes the host for a protective response
against a lethal respiratory viral infection. In addition, it has been
suggested that p80 functions as a competitive antagonist of IL-12
p70. Mouse Con A-activated splenocytes display identical binding
affinities for p80 and IL-12, and in these cells p80 competitively inhibited IL-12 binding and IL-12-dependent proliferation. Furthermore, p80 is able to inhibit IL-12-dependent IFN-γ production in
freshly isolated splenocytes.

Mouse IL-15
Interleukin-15 (IL-15) was discovered in the supernatant of Simian
kidney epithelial cell line CV-1/EBNA, as a soluble factor capable
of supporting proliferation of the IL-2-dependent cell line, CTLL2. IL-15 is a regulatory cytokine, produced by dendritic cells, epithelial cells, fibroblasts, and monocytes. IL-15 plays an important
role in immune response and shares many functions with IL-2.
For example, it stimulates the proliferation of activated T cells, NK
cells, and B cells, and induces immunoglobulin synthesis by B cells
stimulated by anti-IgM or CD40 ligand. In addition, IL-15 promotes
the development of dendritic cells and induces the production
of proinflammatory cytokines from macrophages. IL-15 acts as a
bridge between innate and adaptive immunity because of its diverse roles in the immune system.
IL-15 binds to heterotrimeric receptors composed of IL-15Rα, IL15Rβ, and IL-15Rγc. IL-15 shares the receptor chains β and γc with
IL-2. IL-15 is normally not secreted in soluble form but is instead
held on the cell surface bound to a unique receptor, IL-15Rα, especially on dendritic cells. Cell-bound IL-15 is then presented in
trans form to T cells and NK cells and is recognized by the γc receptor on these cells; such recognition maintains cell survival and

Interleukin-17A (IL-17A) is the founding member of the IL-17 family, a group of six structurally related pro-inflammatory cytokines.
IL-17A, secreted by activated CD4+ Th17 cell subpopulation, elicits multiple biological activities on a variety of cells including:

Interleukin-17A/F (IL-17A/F) is part of the IL-17 cytokine family
which consists of six structurally related proteins (IL-17A, B, C, D,
E, and F). IL-17A is expressed primarily by Th17 cells, a subset of
CD4 T cells. IL-17F is most closely related to IL-17A. The two molecules share 50% amino acid sequence homology. Like IL-17A, IL17F mRNA and protein have been detected in Th17 cells. IL-17F

Mouse IL-17F
Interleukin-17F (IL-17F) belongs to the IL-17 cytokine family which
includes IL-17A, B, C, D, and E (also called IL-25). IL-17F shares the
strongest homology to IL-17A. They share 50% amino acid sequence homology. Recently, an IL-17–IL-17F heterodimer was
found to be expressed in Th17 cells, together with IL-17 and IL17F homodimers. IL-17 and IL-17F gene promoters share the same
pattern of chromatin remodeling in differentiated Th17 cells. T cell
populations in vitro and in vivo have shown different ratios of IL-17
and IL-17F expression. IL-17, IL-17F regulates proinflammatory

IL-17F and IL-17A were involved in host defense against mucoepithelial infection by Staphylococcus aureus and Citrobacter rodentium. Additional studies in vivo have suggested a role for IL-17F in
the induction of neutrophilia in the lungs and in the exacerbation
of Ag-induced pulmonary inflammation.

Interleukin-25 (IL-25), also known as IL-17E, is a member of the IL-17

Interleukin IL-27 is a heterodimeric cytokine consisting of EBV-induced gene-3 (EBI3, an IL-12 P40-related protein) and P28 (a newly
discovered IL-12 P35-related protein). It is a member of the IL-6/
IL-12 cytokine family and mainly produced by antigen-presenting cells, including macrophages and dendritic cells. IL-27 acts on
T cells and NK cells. It has been reported that IL-27 drives rapid
clonal expansion of naïve CD4+ T cells, promotes Th1 polarization,

Interleukin-33 (IL-33) belongs to the IL-1 family and is closely related in structure to IL-18 and IL-1b. These cytokines are synthesized as biologically inactive precursor and are cleaved by the

Mouse IL-34 shares a sequence identity of 71% with human IL-34
on the amino acid level, and the IL-34 gene is syntenic in the
human, chimpanzee, rat, and mouse genomes. IL-34 has no apparent consensus structural domain or motif and does not share
sequence homology with M-CSF; nevertheless, it binds to the

ligand interactor (TACI) is a TNFR family member molecule and
part of the so-called BAFF system. This includes two ligands: B

Interleukin-17A (IL-17A) is the founding member of the IL-17 family, a group of six structurally related pro-inflammatory cytokines.

Interleukin-17F (IL-17F) is one of the six IL-17 family members (IL17A through IL-17F), that shares the closest homology (~50%
amino acid identity) to IL-17A. Like IL-17A, IL-17F has also been associated to autoimmune diseases. IL-17F is produced and secreted
by Th17 cells and activated monocytes. Like IL-17A, IL-17F has also
been linked to multiple biological activities on a variety of cells
including: inhibition of angiogenesis, induction of IL-2, IL-6, IL-8,
G-CSF, GM-CSF, GROα, ENA78, TGFβ, MCP1, and ICAM1 production in epithelial cells, endothelial cells, and fibroblasts. IL-17F and

Human IL-34 shares a sequence identity of 99.6%, 72% and 71%
with chimpanzee, rat, and mouse IL-34, respectively, on the amino
acid level. The IL-34 gene is syntenic in the human, chimpanzee,
rat, and mouse genomes. IL-34 has no apparent consensus structural domain or motif, and does not share sequence homology

RANTES is a 7.8 kD member of the CC family of chemokines also
known as CCL5, which is expressed in T-cells and monocytes/macrophages. RANTES is strongly chemotactic for monocytes/macrophages, memory T-cells, and eosinophils, and induces histamine

PDGF-AA, AB, and BB dimers are processed intracellularly and secreted as active dimers that readily activate PDGF receptors (PDGFRs).

TGF-β1 is synthesized in cells as a 390-amino acid protein. Furin
cleaves the protein at residue 278, yielding an N-terminal cleavage product which corresponds to the latency-associated peptide
(LAP) and the 25-kD C-terminal portion of the precursor constitutes the mature TGF-β1. In addition to furin, some other activators can release TGF-β from LAP. TGF-β activators include proteases that degrade LAP, thrombospondin-1, reactive oxygen

TGF-β1 seems to have dual effects on human Th17 differentiation
in a dose-dependent manner. While TGF-β1 is required for the expression of RORγt, in human naive CD4+ T cells from cord blood,
TGF-β1 can inhibit the function of RORγt at high doses. By using

14 (Fn14)/TWEAKR has been described as a receptor for TWEAK,
and it is associated with proliferation of endothelial cells and angiogenesis. However, TWEAK mediates signal transduction and
linear differentiation of monocyte/macrophage cells lacking Fn14/
TWEAKR, suggesting that such cells contain an alternative TWEAK
receptor. Elevated levels of TWEAK and/or Fn14 have been found
to be associated with the pathogenesis of rheumatoid arthritis,
skeletal muscle wasting, systemic lupus erythematosus, multiple
sclerosis, stroke, neuroinflammation and neurodegeneration, and
several types of cancer. The pathological functions of TWEAK are
primarily attributed to its ability to induce the expression of several proinflammatory cytokines, chemokines, cell adhesion molecules, and matrix-degrading enzymes mainly through the activation of NF-B, a major proinflammatory transcription factor. More
recently, it has been described that CD163 (a scavenger receptor)
might act as a receptor “decoy” for the ligand TWEAK.

Recombinant human VEGF-165, amino acids Ala27-Arg191 (Accession# NM_001171626) was expressed in E. coli. This protein
was purified to greater than 95% purity.
Bioactive Protein: This VEGF-165 protein is biologically active
with an ED50 = 2.5 to 4.5 ng/mL, corresponding to a specific
activity of 2.2–4 × 105 units/mg, as determined by HUVEC
cell proliferation in a dose dependent manner. The protein
is provided lyophilized, and endotoxin is less than 0.1 EU/µg
(<0.01 ng/µg).

VEGF-165 plays a critical role in new blood vessel formation in
vivo. VEGF-165 induces angiogenesis on different levels: it acts
as mitogen especially on endothelial cells, raises the vessel permeability and dilation by releasing NO, and has chemotactic impact on monocytes/macrophages, which plays a crucial role in
inducing inflammatory neovascularization. Angiogenic activities
of VEGFs are mediated primarily through two receptors, VEGFR-1
and VEGFR-2. VEGFR-2 signaling is enhanced by interactions with
co-receptors such as heparin/heparan sulfate and Neuropilin 1
(NRP-1). The expression of VEGF-165 in the tumor microenvironment influences the differentiation of bone marrow-derived pericytes to the Ewing’s sarcoma vasculature. VEGF induces osteoclast
migration and activation. In addition, it supports the survival of
mature osteoclasts. These processes play a key role in repairing
and remodeling bone during bone development. VEGF is upregulated by hypoxia and by overexpression of oncogenes such as
mutant ras and c-myc in tumours. Immobilized forms of perlecan
and PlnDI (inmovilized and soluble) bind VEGF-165 to coordinate
developmental angiogenesis modulating VEGF-165/VEGFR-2 signaling. In addition, CXCL8/IL-8 stimulates VEGF expression and
VEGFR2 activation in endothelial cells.

than 98% purity.
Bioactive Protein: This TWEAK protein is biologically active
with an ED50 < 10 ng/mL, corresponding to a specific activity
of > 1 × 107 units/mg, as determined by the dose dependent
stimulation of production of IL-8 by human PBMC. Bottled
lyophilized, and endotoxin is less than 0.1 EU/µg (<0.01 ng/µg).
