No Pain, No Gain: Understanding Muscle Pain Jankowski Lab – Anesthesia/Pain Management Jordan Lamb Project Description o Goal – Better understand molecular mechanisms that lead to muscle pain after injury in order to determine better treatments for musculoskeletal pain o Muscle-specific injury model in mice o Block blood flow in right forepaw by tying off brachial artery (Ischemia) o Remove blockage after several hours to allow blood flow again (reperfusion) o Causes changes in the muscle tissue and in gene expression in afferent neurons that carry pain input from muscle to CNS Project Description Cont. o Three types of procedures to study pain produced after injury o Behavior – guarding, grip strength, threshold they respond to mechanical stimuli o Electrophysiological recording – responsiveness to mechanical, thermal, and chemical muscle stimulation o Molecular Assays o My role o Focus on molecular assays o Real-time PCR analysis What? So What? Now What? o Basic laboratory skills – pipetting, weighing chemicals, preparing solutions, mouse handling, etc. o Exposed to variety of different procedures o RNA isolation, protein isolation, reverse transcription, PCR, cell culture o Assist with animal surgery o About myself o Like working in structured environment – following procedure o Gaining independence o Asking for help What? So What? Now What? o Bettering understanding of neuroscience o Classroom and daily life o Chance to explore other science/medicine related careers o Perceptions on research process o A LOT of work o More respect o Prepared to continue research this summer and in the future
