Description
Histones are basic nuclear proteins that are responsible for the nucleosome
structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of
approximately 146bp of DNA wrapped around a histone octamer composed
of pairs of each of the four core histones (H2A, H2B, H3, and H4) limiting
DNA accessibility to the cellular machineries which require DNA as a
template. Histones play a central role in transcription regulation, DNA
repair, DNA replication and chromosomal stability.
Histone H3 is a 17 kD nuclear protein. The N-terminal tail of histone H3
protrudes from the globular nucleosome core and can undergo several
different types of post-translational modification in different physical status.
These modifications include the covalent attachment of methyl
(methylation) or acetyl (acetylation) groups to lysine and arginine amino
acids and the phosphorylation of serine or threonine. Di- and Trimethylation of Lysine 9 are associated with gene repression and
heterochromatin, while mono-methylation of K4 and acetylation of lysine14
are associated with active genes. Histone H3 has been shown to interact with
Rsk-2, MSK1, GCN5 family of HATs, HDACs, PARP, SET7/9, and CARM1.

Chromatin Immunoprecipitation (ChIP) was
performed using commercial Protein-G
coated 96 well high-throughput ChIP assay
kit by loading 3 µg of cross-linked chromatin
samples from HeLa cells with either A) 1:100
dilution of Go-ChIP-Grade™ Purified antiHistone H3 (C-terminus) Antibody (clone 1B1B2), B) equal amount of Purified Mouse
IgG3, κ Isotype Control Antibody (clone MG335), or C) competitor's ChIP-grade Purified
anti-Histone H3 Antibody and D) equal
amount of matched Isotype Control Antibody
as recommended by the manufacturer. The
enriched DNA was purified and quantified by
real-time qPCR using primers targeting
human GAPDH gene region. The amount of
immunoprecipitated DNA in each sample is
represented as signal relative to the 5% of
total amount of input chromatin.

Immunogen
This monoclonal antibody was raised against a peptide sequence derived from the C-terminal
domain of human Histone H3 protein.
Formulation
Phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide.
Preparation
The antibody was purified by affinity chromatography.
Concentration
0.5 mg/ml

The antibody solution should be stored undiluted between 2°C and 8°C.

Recommended Usage
Each lot of this antibody is quality control tested by ChIP Assay. The suggested dilution for ChIP
application is 1:50-1:150 by volume. For immunohistochemical staining on formalin-fixed
paraffin-embedded tissue sections, the suggested dilution is 1:1000. It is recommended that the
reagent be titrated for optimal performance for each application.
Application Notes
25 µg and 100 µg of Go-ChIP-Grade™ Purified Antibody can be used for 2-7 or 10-30
immunoprecipitations, respectively, at the recommended dilutions.

form a tetramer. This tetramer further combines with two H2a-H2b dimers to form the compact
