Bacteria cells have plasmids, much smaller than bacterial chromosome!
Things to remember about plasmids:
1. During replication, the plasmids must also replicate.
2. Plasmids can be removed and harvested from the cell.
3. Plasmids are used in genetic engineering and biotechnology.
Plasmid structure:
1. DNA is antiparallel
2. Held together by hydrogen bonds
3. Plasmids have a small # of genes

Genetic Engineering
1. Goal: place desired gene into a plasmid
2. Cut both gene of interest and plasmid with restriction enzymes
3. Leave sticky ends that will adhere to each other
4. Hydrogen bonds will form between sticky ends
5. Ligase will create phosphodiester bonds between the bases.
