However, without its cell wall, the naked protoplast is spherical and is not capable of dividing, growing, or becoming specialized for function. Molecules of the cell wall provide mechanical strength, regulate porosity, and control cell-cell adhesion. The functions of the wall are not only mechanical but also biological. Like the animal extracellular matrix, plant cell walls are a source of signaling molecules that elicit a range of cell behaviors, committing the cell to particular developmental programs. The goal of my research is to understand how the molecular machinery of the plant cell wall contributes to cell growth and specialization, and thus to the final stature and form of plants. The plant cell wall is a highly organized composite of many different polysaccharides, proteins, and aromatic substances that undergo dynamic changes during cell division, expansion and differentiation. However, it has been difficult to ascribe specific functions to these molecules. As part of an NSF-funded genomics project (see http://cellwall.genomics.purdue.edu), we have used a high-through-put infrared spectroscopic screen to identify a broad-based collection of mutant plants with variation in cell wall amount, composition or molecular architecture. In this Purdue-centered collaboration with Dr Nicholas Carpita, Department of Botany and Plant Pathology, we have screened populations of over 900 homozygous cell-wall-related mutant lines of Arabidopsis, about one-quarter of which have alterations in cell wall composition or architecture. We are using these materials to define the relationship between genetically-defined changes in plant cell wall-related proteins (biosynthetic, hydrolytic, and structural) and cell wall molecular architecture and structural properties and to examine the downstream consequences for cell growth and differentiation. Both final cell size and rate of cell expansion are constrained by the cell wall. I have proposed that the wall-loosening mechanisms that permit cell growth to occur are regulated by the biophysical properties of a network of pectic polysaccharides. Pectins are some of the most structurally complex biomolecules known, comprise 20 to 50% of the mass of dicotyledonous cell walls, and are cross-linked together and to other cell wall molecules in a variety of ways. In order to elucidate the role of pectin in cell elongation, my lab uses mutants and transgenic approaches to generate Arabidopsis plants of altered pectic composition. For example, we have identified a 7-member gene family in Arabidopsis, the MYST gene family, that encode rhamnogalacturonan (RG) lyases, an enzyme activity more commonly associated with plant pathogens for cell wall hydrolysis. RG-lyases are enzymes that specifically recognize and cleave glycosidic bonds between L-rhamnose and D-galacturonic acid residues in the backbone of RG-I molecules. Mutation and over-expression of MYST gene family members each have dramatic impacts on plant growth. Our data indicate crucial functions for MYST proteins and their substrate in plant development. 3 Plant cell walls are the source of lignocellulosic biomass, an untapped and sustainable resource for biofuels production. We require tremendous increases in agricultural productivity to meet the competing demands of food, feed and now biofuels without increasing our agronomic footprint. Using a toolkit of plant genetics and molecular biology, we aim to optimize dedicated bioenergy crops by gaining genetic control of the amount, composition and structure of cell walls for their end-use as feedstocks in production of energy-rich biofuels. A frontier in energy research is to develop technologies that maximize the energy and carbon efficiencies of biofuel production by the rational and synergistic design of both physical and chemical conversion processes and the biomass itself. I lead an Energy Frontier Research Center focused on the direct catalytic conversion of biomass to biofuels, and I am building an interdisciplinary program to tailor biomass for its end-use in thermal or catalytic conversion pathways for energy-rich fuels and other bio-based products.
