Purified Sp1 protein Catalog No: 31137 Expressed In: HeLa cells Quantity: 2 µg Source: Human Buffer Contents: 2 µg of Purified Sp1 protein and Dilution Buffer AM1 (20 mM TrisCl (pH 8), 20% glycerol, 100 mM KCl, 1 mM DTT and 0.2 mM EDTA). Protein is supplied at 0.08 µg/µl in Dilution Buffer AM1. Background: Sp1 (specificity protein 1) is a human transcription factor involved in gene expression in the early development of an organism. The protein is 785 amino acids long, with a molecular weight of 81 kDa. The SP1 transcription factor contains a zinc finger protein motif, by which it binds directly to DNA and enhances gene transcription. Its zinc fingers are of the Cys2/His2 type and bind the consensus sequence 5′-(G/T)GGGCGG(G/A)(G/A)(C/T)-3′ (GC box element). It was discovered in 1983 and has since been modified to form Sp1C, which has a zinc finger protein DNA binding domain. Protein Details: The natural Sp1 fraction (accession number NM 138473) was purified from HeLa cell nuclear extract by using wheat germ agglutinin affinity chromatography. The purified protein is greater than 95% homogeneous and contains no detectable protease, DNase or RNase activity. Application Notes: Purified Sp1 is suitable for in vitro transcription, DNase I protection and gelshift assays. 50-100 ng is sufficient for reconstituted transcription assays and 5-25 ng in a 20 µl volume are sufficient for gelshift assays. The molecular weight of the protein is ~105 kDa. This protein is not ® recommended for TransAM assays. NOTE: The presence of Poly [d(I-C)] in buffers may affect protein functionality and should be avoided. Storage and Guarantee: This product is guaranteed for 6 months from date of receipt. This product is for research use only and is not for use in diagnostic procedures. North America 877 222 9543 • Europe +32 (0)2 653 0001 • Japan +81 (0)3 5225 3638 • www.activemotif.com
