HMB is converted in the body to HMG­CoA, a molecule that is important for
sarcolemmal integrity. The sarcolemma is a specialised cell membrane found
around muscle fibres. By protecting this membrane HMB can decrease
damage to muscle fibres and speed up the recovery period. Scientific studies
also show that HMB decreases markers of muscle damage after exercise.

and Protein Synthesis
Scientists now think that HMB can improve muscle gain and prevent muscle
loss through three different pathways. The first of these is to stimulate protein
synthesis by signalling through the mTOR pathway, just the way leucine does.
What HMB is doing in effect is to tell cells that there are a lot of nutrients
available so they can go ahead, make a lot of new proteins and lay down new
muscle fibres.

It is also thought that HMB directly inhibits protein degradation through a
pathway known as ubiquitination. In this pathway a small molecule called
ubiquitin (because it is found in all cell types) is linked to protein molecules.
This targets them for degradation in proteasomes. Obviously if you are a body
builder or an athlete you don’t want your proteins to be degraded. HMB inhibits
protein ubiquitination, and therefore has anti­catabolic activity.
These three very different mechanisms can add up to a whopping 18%
increase in performance and muscle gain as discovered by scientists in
clinical studies.
Scientists have known for a long time that the BCAA amino acid can inhibit
protein degradation, among its other important benefits for muscle gain.
However the does of leucine required for this anti­catabolic effect was much
higher than for its anabolic effects. The dose was in fact much higher than that
found in BCAA supplements. It is now thought that this anti­catabolic effect is
due to HMB.
If you are already taking a BCAA supplement you might question whether you
also need to buy HMB. The answer is yes. Although leucine, one of the
branched chain amino acids is the parent coupound, only a small fraction,
around 5% is degraded to HMB. The vast majority of leucine gives rise
isovaleryl­CoA. It is estimated that you would need to consume 60g of leucine
to get 3g of HMB. Obviously that is not practical, and you need to take an HMB
supplement directly to obtain the anti­catabolic effects.
