Read over the handout on ESTIMATING SIZES OF DNA BANDS BY SEMI-LOG PLOT (Science
Education Partnership. Pay careful attention to pages 1, 2 and 4.

Now, Read the rest of the information on S120 – S121 in Big Idea #3 – Genetics Information
and Transfer – Biotech Restriction Enzyme Analysis of DNA.

Using this data, complete the exercise as directed. Perform the appropriate
measurements for all fragments on the Ideal Gel on page S121n and record on the
chart in this handout.

Using the data for fragment HINDIII, plot your line of ‘Best fit’ on the semi-log paper
blank provided for you with the handout in II. Make the Best Fit line RED.
Determine the BP lengths for the other fragments using your graph and fill in the
rest of your chart.

