1. Overview
Cell surface adhesion molecules play vital roles in numerous cellular processes. Some of these
include: cell growth, differentiation, embryogenesis, immune cell transmigration and response, and
cancer metastasis. Adhesion molecules are also capable of transmitting information from the
extracellular matrix to the cell.
There are four major families of cell adhesion molecules:
 Immunoglobulin (Ig) superfamily cell adhesion molecules (CAMs)
 Integrins
 Cadherins
 Selectins
2. Immunoglobulin (Ig) superfamily Cell Adhesion Molecules (CAMs)
A. Intercellular Adhesion Molecule-1 (ICAM-1)
ICAM-1 expression is weak on leukocytes, epithelial and resting endothelial cells, as well as some other cell
types, but expression can be stimulated by IFN-gamma, TNF-alpha, IL-1 beta and LPS.
ICAM-1 seems to be the initial marker of inflammatory reactions and is expressed prior to, and to a greater
extent than is HLA-DR.

B. Vascular Cell Adhesion Molecule-1 (VCAM-1)
Human VCAM-1 (CD106), a member of the immunoglobulin superfamily, is a cell surface protein
expressed by activated endothelial cells and certain leukocytes such as macrophages. VCAM-1 expression is
induced by IL-1beta, IL-4, TNF-alpha and IFN-gamma

3. Selectins
1. Introduction
Selectins mediates the initial interactions of leukocytes and platelets with
endothelial cells. They guide non-activated polymorphonuclear cells to the
areas of inflammation in creating first, loose contacts with the endothelial
layer.
2. Clinical Applications of Selectins
sE-Selectin
Allergic reactions, ocular diseases, septic shock, vascular infection and
inflammation, graft-versus-host-disease
sL-Selectin
Allergy, Deep venous thrombosis, Diabetes mellitus, Kawasaki Syndrome,
neonatal bacterial infection, sepsis

Adult respiratory distress syndrome acute lung injury, Ischemia-reperfusion
