Biophysical methods are essential tools during
the entire drug discovery process – from early
stage R&D through to QC and manufacturing.
They can be used for optimization of protein
constructs and verification of proper and
consistent protein folding, identification of
optimal assay buffer, screening of fragments
and the confirmation of binding to the right or
allosteric binding site(s) of the target
molecule. You can also measure binding
affinity, thermodynamics and kinetics to be
able to perform enthalpy and residence time
based optimization of leads into candidate
drugs. In this seminar we will provide an
introduction to label free techniques:
Differential Scanning Calorimetry (DSC),
Surface Plasmon Resonance (SPR) and
Isothermal Titration Calorimetry (ITC) and
highlight their use in a several application
areas in and around tyrosine kinases (VEGF,
EGFR) and transmembrane receptors (BACE1,
CXCR4 etc).
