Description: Recombinant monoclonal antibody to Thyrotropin receptor. Manufactured using
AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma K1-70. This
antibody was created using our proprietary Fc Silent™ engineered Fc domain containing key point
mutations that abrogate binding of Fc Receptor and complement.

Immunogen: K1-70 was prepared by creating a hybridoma cell line using lymphocytes from a
patient with hypothyroidism and high levels of TSHR autoantibodies. The binding epitope on the
TSHR is likely ot be closely related to the TSHR ligand binding region due to the ability of unlabelled
K1-70 to inhibit binding of labelled TSH.
Specificity: K1-70 binds specifically to the ectodomain of TSHR. This antibody was derived from a
patient with hypothyroidism in which autoantibodies to TSHR which having blocking activity are
produced. TSHR is a 7-transmembrane G protein-coupled receptor found primarily on the suface if
thyroid epithelial cells which binds to its ligand TSH and plays a central role in controllingt thyroid
cell metabolism. Stimulation of TSHR by TSH results in a signal cascade which activates adenylyl
cyclase, resulting in an increase in intracellular [cAMP] - cAMP activates iodine pumping,
thyroglobulin synthesis, iodination, endocytosis and proteolysis, thyoid peroxidase activity and
hormone release within the thyroid cell.
Application Notes: K1-70 can block stimulation of TSHR by TSH, K1-18, other thyroid stimulating
MAbs and patient serum stimulating type TSHR autoantibodies. The antibody has no stimilatory
activity of its own - no cAMP is produced. The production of cAMP in TSHR transfected CHO cells.
