Incaseofhealthyandfreshplantswhicharenotovergrownwithalgae,protoplasmstreamingwillsoonbecomeobservable.
Shouldthelearninggrouphavefailedtoproduceasufficientnumberofdrawings,thecellsofElodeamayalsobedrawn.Itis
clearlyvisiblethatalargepartofthecellisapparentlyempty(vacuole)andthatthechloroplastsoccuronlyinasmalldelimited
area(cytoplasm).However,thedynamicsofthisprocesscannotberecordedinadrawingorbymeansofphotographybutshall
bedescribedinwords(3).
ad3.Observation
Thestudentsaresupposedtoidentifyandbeabletodescribethemovementofthecytoplasmasrotational.Forexample,the
cytoplasmmovesaroundthevacuoleinacircularmotion,rotatinginaclockwisefashion,orsimilar.

Methodsandobservations
Elodea(waterthyme,waterweed)isaplantwhichisveryabundantinwatersofNorthAmericaandEuropeandistherefore
referredtoas"waterpest"inseveralcountries,e.g.Germany.Surely,youmusthavealreadyseenthisplantinanaquarium.It
qualifiesfortheseexperimentsbecauseithasverythinleaves.
1.Preparationofslides
Pipetteonedropofwaterontoaslide.
Pickoffaleafletfromtheupperpartoftheplant.Olderleavesareovergrownwithalgaeandshouldthereforenotbeused.
Theleafletisplaceddirectlyintothewaterdropandcoveredwithacoverslip.
2.Microscopy
Withoutpriorstaining,thecell’sprotoplasm(cytoplasm)appearsratherpaleandisdifficulttoobserveforthisreason.Mostof
thespacewithinacellisoccupiedbythevacuoleanyway.Thesmallstripofcytoplasmcontainsmanygreenchloroplastswhich
apparentlyflowpassivelywiththestream.Thisallowsustofollowthephenomenonofprotoplasmstreamingindirectly.
Viewthespecimenwithincreasingpowers.
Ifpossible,examineleafareasinwhichthereisonlyonelayerofcells.Thisisthecaseinboundaryareasandalongthe
midveinoftheleaflet.
Youmusttakeyourtimeforthisobservation.Streamingwilloftenbevisibleseveralminutesafterthemetabolicprocesses
havebeenactivatedasaresultofillumination.
