Homework 4:
Due in your lab starting September 30th and will be grading by your Lab TA.
6pts (all or none):
Q1: Draw a gene with six exons on the line below.
The coding region (ORF) is spread out amongst 5 exons.
Two of those exons contain 5’UTRs and two of those exons contain 3’UTRs.
In addition, the last exon is composed only of 3’UTR sequence and contains the
only polyA recognition site sequence that will end up in all mRNA species.
TSS (all regions)
5’UTR (all regions)
Coding sequences (all regions)
3’UTR (all regions)
AATAAA sequence
5’ Donor site (all of them)
3’ Acceptor site (all of them)

Q2: The sequence below represents a stretch of amino acids in a given
polypeptide.
NILAESRTQDF
4pts. IF THEY GET 8 CORRECT, THEN GIVE FULL CREDIT
Write out an analogous sequence (maintaining the above order) with all
amino acids being different from above. You are designing a homologous
protein.
_QLIVDTKSNE (Y or W)__
For I L A residues
(any of the following residues would be acceptable I L A G V)
IF THEY GET 8 CORRECT, THEN GIVE FULL CREDIT
4pts. Amino acid comparisons between TAS2R38 from

Human vs. Chimp TAS2R38 (top amino acids vs. bottom). Middle amino acid
layer represent identities (Identical residues). A plus sign (+) represents
similarities. A blank space represents no similarities.

Which is more Identical: Human vs. Chimp or Mouse vs. Rat___Human and.
Chimp____,
and provide the numbers that give rise to your conclusion__98% identity vs.
88%____.
Which is more similar: Human vs. Chimp or Mouse vs. Rat__ Human and
Chimp____,
and provide the numbers that give rise to your conclusion __98% similarities
vs. 94%____.
Underline the Human vs. Mouse identities that exist for at least 10 consecutive
amino acids. See figure
Is Rat or Mouse TAS2R38 closer to the Human sequence and why?
__Either Rat or Mouse is acceptable_____
However Mouse is more homologous to Human in the underlined regions
(Question 3), but Rat has 266/332 similarities to Human versus 265/332 for
Mouse.
