2002
Completion of doctoral thesis. This project involved the use of an innovative Left
Ventricular Assist Device (Paraaortic Counterpulsation Pump) during cardiogenic
shock, in a large animal (pig) experimental model. At the same time, I started
experimental research protocols in the University of Athens using bone marrow
derived stem cells for amelioration of ventricular remodeling.
2004-2005
Research and training in stem cells and tissue engineering, in Imperial College
London. I was involved in the development of a tissue engineered cardiac valve
using biomaterial scaffolds and mesenchymal stem cells.
2005-2009
Basic and translational research using cardiosphere-derived stem cells for
myocardial regeneration in Johns Hopkins University and Cedars Sinai Medical
Center.
I was involved in the development of imaging methods, both for small and large
animal models, aiming at cell tracking after delivery in the heart. Our purpose was
to investigate the efficiency of the delivery methods and study short term cell
retention and long term engraftment. With the help and support of scientists from
basic research disciplines such as molecular biology and physics, I applied every
available modality (bioluminescence imaging, Single Photon Emission Tomography
and Positron Emission Tomography, Magnetic Resonance Imaging) in our research
and defined the exact retention and survival rates of CDCs in the myocardium. I
developed protocols for direct radiolabeling of CDCs that allowed cell tracking using
the relevant modalities. In addition, we used reporter gene technology for
bioluminescence, SPECT and PET imaging. We were the first to provide compelling
evidence that a human gene (sodium-iodide-symporter) can be used as a reporter
gene for tracking genetically labeled cells by SPECT and PET imaging (Young
Investigator Award in the 2007 American Heart Association Sessions). This is an
important development considering the clinical translatability of this method. Using
the same modalities, we were able to apply simple techniques to improve cell
retention and in parallel the functional benefit of CDC therapy. These studies were
financed by NIH grants, as well as by other distinguished funding agencies
(Reynolds foundation and WW Smith foundation).
2009-present
I returned to Greece with an appointment in the University of Athens, School of
Medicine, in 2009. I established a new, basic research laboratory, as an addition
to the pre-existing laboratory for experimental research in large animals, in the 3rd
Department of Cardiology. The new laboratory has tissue culture, molecular biology
and small animal surgery capabilities. Currently, three MDs with experience in basic
research are performing experiments in this lab. In addition, three medical
graduates are conducting research in this lab, as a part of their doctoral thesis (with
myself as the supervisor). Several experimental studies involving the use of
cardiosphere-derived stem cells (both autologous and allogeneic) are being
conducted, currently in this lab.
In addition, I designed a randomized clinical study using peripheral blood
mononuclear cells (after stimulation with growth factors) for prevention of left
ventricular remodeling, in patients with large anterior myocardial infarcts. The trial
was conducted in the Dept. of Therapeutics of the University of Athens and
preliminary results were presented in American Heart Association Scientific
Sessions (oral presentation-November 2011, Orlando, USA). Final results are
available and a manuscript is under preparation.
At the same time I continued my collaboration with Cedars Sinai Medical Center.
Among other projects, I was the primary author of the protocol for a clinical trial
(ALLSTAR- NCT01458405), where allogeneic cardiosphere-derived stem cells will
be administered to patients, by intracoronary delivery, in the setting of chronic MI
(1month to 1 year post MI). This trial has recently started enrolling patients.
