The efficiency of this process will be highly improved if we can reveal the negative characteristic of the compounds in the early phases, as the HTS. This issue could be tackled by developing more in vivo-like assays. The Primary cells are extremely difficult in culture and have a limited expansion potential. The use of stem cells is one of the strategies to create better cell models closer to the cells in human target tissues and more predictive for efficacy and safety in vivo than the stable cell lines currently used. The adult stem cells could be the key as they can expand to high number of cells and differentiate into any adult specialized cell type. At the same time, the use of adult stem cells overcomes any ethical consideration. But because the telomeres shortening restrict their ability to proliferate we carried out an immortalization process to increase their expansion potential. We immortalized adult mesenchymal stromal cells from adipose tissue (ADSCells) and most of the obtained clones are able to differentiate into different types of primary cells and it is easy to scale them up for uHTS requirements.
