Synthetic peptide from human retinoblastoma protein containing phosphorylated serine 608.
SPECIFICITY
This antibody reacts with retinoblastoma protein phosphorylated at serine 608. There is no cross reactivity with
unphosphorylated retinoblastoma protein.
REACTIVITY
Reactivity of this antibody was confirmed using human NALM-6 (pre B acute lymphoblastic leukemia) cells nuclear extract.

USAGE
Working concentrations for specific applications should be determined by the investigator. Appropriate concentrations will be
affected by several factors, including secondary antibody affinity, antigen concentration, sensitivity of detection method,
temperature and length of incubations, etc. The suitability of this antibody for applications other than those listed below has
not been determined. The following concentration ranges are recommended starting points for this product.
ELISA: 0.1-1.0 µg/ml
Western Blotting: 1-5 µg/ml
Immunoprecipitation: 10 µg/IP reaction
STORAGE
Store at 2-8°C for up to one month. Store at –20°C for long term storage. Avoid repeated freezing and thawing.

BACKGROUND
Entry into S phase from late G1 is controlled by phosphorylation of the retinoblastoma protein (Rb). Mutations affecting Rb or
proteins that control Rb phosphorylation are frequent in human tumors. The cell cycle distribution of childhood acute
lymphoblastic leukemia (ALL) cells shares many features with that of normal CD34+ progenitor cells. The majority of CD34+
and ALL cells reside in G1 phase although their cell cycle distribution within the G1 phase may be different. Entry of the cell
into late G1 phase is controlled by phosphorylation of Rb. Different phosphoforms of Rb were recognized by their slower
mobility on SDS-PAGE in ALL cells vs. CD34+ cells. In ALL cells, Rb was phosphorylated at ser-608, ser-780 and ser-795
unlike CD34+ cells. This evidence suggests that G1 phase distribution of ALL and CD34+ cells is different and that ALL cells
progress to mid G1 phase while unstimulated CD34+ cells reside in early G1 phase.
