Supplementary Figure 1. HPLC purification and Mass Spectrometry analysis of WE-14 and
ChgA 29-42 peptides. The peptides were synthesized and purified as described in Material and
Methods section and analyzed at the University of Western Ontario (UWO) Biological Mass
Spectrometry Laboratory.
Supplementary Figure 2. ChgA29-42 primed BDC2.5 T cells express activation surface
markers. Spleens were harvested from 6-weeks-old BDC2.5 NOD mice and cells were cultured
in 96-well plates at 2 x 105 cells/well in the presence of 50 μg/ml of ChgA29-42, WE14, PS3 and
negative control peptides in culture media at 37°C for 72 h. Cell were surface stained with
fluorochrome–conjugated antibodies to CD4, CD69, CD44 and CD25. The results for activation
markers (thick line for antibodies and thin line for the isotype controls) are shown on gated CD4+
T cells.
