REVIEW:
Do we use introns or exons?
Draw a strand of DNA that contains silencer, repressor, basal transcription factors, TATA box,
(transcription factors):

Why does these processes need to happen?
Four advantages to use plasmids in gene cloning:
1.
2.
3.
4.
Plasmids are self ____________.
They are ___________________ for cell survival.
They can ______________ into the bacterial genome
They contain antibiotic _____________ genes.

Recipe for gene cloning:
What do we need?
Step 1:

Step 4:
What have we made? What can we use it for?
Looking ahead: Gel Electrophoresis
1. You leave for the bathroom and coming back, you notice that someone has taken a bite from
your cookie you left on your plate. You collect the saliva from the crime scene. You carry
out PCR amplification and electrophorese the DNA. You suspect your mom, dad or your
little brother (suspects 1, 2, and 3) to have taken a bite and collect hair samples from them
and carry out the same methods. The results are shown below. Who is guilty of eating your
cookie?
