Objectives:
Molecular Characterization of Pakistani Wheats for Rust Resistance Genes.
Commencement: January 2009
Completion: June 2012
 To detect rusts resistance/susceptible genes of Pakistani wheats using DNA markers.
 To improve rust resistance in Pakistani wheat through Marker Assisted Selection.

Work Done (to date):
 Seeds of 100 wheat genotypes including released Pakistani varieties, advance breeding lines
of Wheat Program and 30 entries included in the Elite Bread Wheat Yield Trials (2008-09)
were acquired from Wheat Program, NARC.
 Genomic DNA of these wheat genotypes was extracted and quantified.
 Presence/absence of Lr-34/Yr-18 of all the genotypes has been determined. This gene cluster
was found in Lyallpur-73, Yecora-70, Punjab-76, Khyber-87, Suleman-96, Saleem-2000,
Panjnad and Fareed-06 and 10 lines included in NUWYT 2006-07.
 Yellow rust resistant gene Yr5 was found in 37 of 60 Pakistani wheat varieties tested.
Yellow rust resistance gene Yr9 was found in 17 varieties. Yellow rust resistance gene Yr10
was present in 42 varieties. Yellow rust resistance gene Yr26 was not found in any of the
variety tested.
 Forty eight Pakistani landraces were tested to determine the presence/absence of stem rust
resistance genes. Stem rust resistance genes Sr24 and Sr31 were not found in any of the
landrace, whereas Sr25 was found in 3 landraces. Forty one landraces had the stem rust
resistance gene Sr2.

 Genomic DNA extracted from 118 Pakistani wheat varieties, which will be screened for the
presence/absence of stem rust resistance genes using 12-15 DNA markers.
 Genomic DNA extracted from 100 Pakistani advanced wheat breeding lines included in
NWDSN and NUWYT (2010-11) for detection of stripe rust resistance genes using 12-15
DNA markers.
11. Significant achievement (if any):
 The presence/absence of major rust resistant gene Lr-34/Yr-18, Yr-5, Yr-9, Yr-10 and Yr-26
determined in 60 commercial varieties and 24 breeding lines. Presence/absence of Sr2, Sr24,
Sr25 and Sr31 determined for 48 wheat landraces.
 15 markers tested for yellow rust resistance genes.
 10 markers tested for stem rust resistance genes.

Work Plan & Methodology:
Amplification of genomic DNA of 100 wheat varieties using 12-15 DNA markers for stem
rust resistance genes
Amplification of genomic DNA of 100 advanced wheat lines using 12-15 DNA markers for
stripe rust resistance genes.
Gel electrophoresis and visualization of PCR products.
Band scoring for determining the presence/absence of genes.
Association analysis of rusts and marker data.
Expected Results/Economic Output:
 Detection of rust resistant/susceptible genes will improve our understanding of the role of
these genes in conferring resistance/susceptibility to prevalent races of rust pathogens in
Pakistan.
 This will aid wheat breeders in pyramiding rust resistance genes into future wheat varieties
through marker assisted selection.
