INTRODUCTION The first report about respiratory infectious bovine rhinotracheitis was published in 1954 (Schroeder and Moys, 1954). The disease was characterised by high fever, agalactia and the symptoms of damage of the respiratory tract. Later the name of the disease was proposed and accepted – infectious bovine rhinotracheitis (Mckercher et al., 1955). The agent of infectious bovine rhinotracheitis (IBR) is the virus of the Varicellovirus genus (BoHV-1) of the Alphaherpesvirinae subfamily of the Herpesviridae family. This is one of the most widespread viruses in the world bovine population due to which many countries engaged in cattle breeding suffer great economic losses due to decreased milk production, fertilisation problems, a loss of bovine younger animals, and abortions caused by the disease. Carriers of this virus could be not only domestic cattle but also wild ruminants (Kendrick et al., 1987, Thiry et al., 1988, Afshar and Eaglesome, 1990, Veselinovič and Medič, 1992, Oirchot et al., 1993, Kaashoek et al., 1996, Hage et al., 1998, Frolich et al., 2002, Frolich et al., 2006). In-depth investigations into both domestic and wild animals carried out showed that the agent of IBR is widespread not only in the groups of domestic ruminants but also among the wild ruminants grazing in freedom (Kalman and Egyed, 2005). The circulation of this virus in the population of wild ruminants can have a great significance to eradication and controlling the virus in the population of domestic animals in Europe. All the more so that transfer of BoHV-1 among different species of animals has not been fully investigated thus far (Lillehaug et al., 2003, Kalman and Egyed, 2005, Frolich et al., 2006). BoHV-1 infection is related to two major syndromes called infectious bovine rhinotracheitis and infectious pustular vulvovaginitis (IPV). The variety of other clinical symptoms is characteristic of the cattle infected with these viruses: conjunctivitis, encephalitis and abortions (Pastoret et al., 1982, Ackermann and Engels, 2006). Also, this virus is an important factor determining the development of a complex of many other diseases especially bovine respiratory diseases (BRD) because BoHV-1 genome codes several albumens, which take part in suppressing a response of the immune system of the organism (Turin et al., 1999, Jones and Chowdhury, 2007, Jones and Chowdhury, 2010). Though after infection with BoHV-1 an immune response of the organism develops, the virus never leaves the organism of the infected animals, and the latter remain latent carriers of this virus for the rest of their life. The protective immunity that forms after the disease is not long-term, and the animals can become sick repeatedly (Mechor et al., 1987, Madic et al., 1995, Babiuk et al., 1996, Schynts et al., 5 2001, Ackermann and Engels, 2006). Stress may cause an acute condition of latent infection therefore viruses can periodically be shed into the environment. Therefore the main source of IBR in the herd are the cattle that had been sick with that disease, which become latent carriers of BoHV-1 and disseminate viruses by means of direct contact or through aerosols, and bulls – with their semen (Vossen et al, 2002, Jones, 2003, Favoreel et al., 2003). BoHV-1 infection is noted for a great variety of clinical symptoms. Since the course of many infections brought about by this virus is subclinical, control measures are to be applied to eradicate the infection itself rather than to suppress the symptoms. Economically IBR is a very important disease. Alongside other dangerous infectious diseases, IBR control is regulated by Part II of Annex E to Directive of the European Union 64/432/EEB. Therefore seeking to eradicate this disease it is recommended to approve national control programmes. BoHV-1 infection, as a cause of the disease, can be diagnosed on the basis of clinical, pathological and epidemiological features. However, to confirm the diagnosis laboratory tests are necessary. In this case complex laboratory diagnostics is based on the detection of the virus itself or its components and specific antibodies to this virus (Parsonson and Snowdon, 1975, Mars et al., 2000). The majority of methods applied in virology are suitable for determining IBR. However, these methods differ in their sensitivity and specificity. Therefore in practice it is expedient to apply different methods that complement one another. Also, it is expedient to make use of the research methods with the help of which it would be possible to investigate biological properties of viruses. This is especially important in assessing IBR prevalence on cattle farms in Lithuania. Until 1996, in Europe, IBR was diagnosed in all the countries in which investigations were carried out; however, the distribution of BoHV-1 was not even. The campaign of eradication this disease was launched in the Scandinavian countries where IBR was diagnosed much rarer. At the present time IBR has been eradicated in Denmark, Sweden, Finland, Austria and in the province of Bolzano in Italy. Germany has achieved much in fighting IBR (Decision of the EEA Joint Committee 2007/584/EB, Nardelli et al., 2008). In Lithuania IBR has been started to be investigated more extensively since 1993. At that time the greatest attention was devoted to eradication of the disease in the herds of bulls – semen producers. By 1997, all seropositive bulls and all infected semen series had been removed from artificial insemination (AI) centres (Šalomskas et al., 1998). Later investigations showed that having implemented these measures bulls of 6 those enterprises were not infected again though during laboratory tests individual seropositive bulls kept under quarantine were periodically detected (Mockeliūnas et al., 2003). However, the IBR condition on industrial dairy farms practically has not been investigated, prevalence of the disease on the national scale has not been analysed. A significant spread of IBR in the herds of dairy cattle in Lithuania, enzooticity of this disease and primary investigations carried out show that much attention should be devoted to the investigation into the epidemiological situation of this disease because only then will it be possible to prepare and apply scientifically substantiated programmes of diagnosing the disease, its prophylaxis, controlling and eradicating it and thus to reduce prevalence of IBR on cattle farms, as well as to minimise economic losses incurred by this disease. Objective of study To investigate the epidemiology of infectious bovine rhinotracheitis in cattle farms of Lithuania, to carry out comparative investigations in the methods of diagnosing IBR, as well as to perform investigations of active and passive immunity and the efficiency of IBR control and prophylaxis. Goals of study 1. To carry out the analysis of prevalence of cattle diseases and causes of the loss of cattle in Lithuania. 2. To establish prevalence of the infectious bovine rhinotracheitis virus on cattle farms in Lithuania. 3. To investigate epidemiology of infectious bovine rhinotracheitis in Lithuania. 4. To carry out a comparative assessment of methods of IBR laboratory diagnostics. 5. To assess the possibilities of the polymerase chain reaction in diagnosing the acute BoHV-1 infection. 6. To carry out investigations into prevalence of IBR and other viral infections (parainfluenza-3, respiratory syncytial virus, bovine viral diarrhoea virus) in cattle population. 7. To evaluate IBR preventative measures taken in Lithuania and their effectiveness. Novelty of study and its scientific value 1. Prevalence of the IBR virus on cattle farms in Lithuania has been determined. 2. The influence of the size of a cattle herd, cattle age and sex on prevalence of IBR has been established. 7 3. The risk of the IBR virus infection frequency according to the distribution of the seropositive cattle has been established. 4. Sensitivity and specificity of the diagnostics method of the enzymelinked immunosorbent assay (ELISA Ab) of antibodies to IBR viruses have been assessed. 5. Possibilities of the use of polymerase chain reaction in diagnosing the acute BoHV-1 infection have been assessed. 6. Peculiarities of prevalence of IBR and other viral infections (parainfluenza-3, respiratory syncytial virus, bovine viral diarrhoea virus) in a cattle population have been compared. 7. The efficiency of IBR control and prophylaxis measures has been assessed. Practical importance of study It has been proved that when ELISA Ab and polymerase chain reaction (PCR) methods are applied together it is possible to ensure a fast and efficient detection of the IBR virus carriers in a cattle herd. It has been established that the PCR method is applied to detect BoHV-1 in clinical and pathological material thus ensuring the confirmation of the diagnosis of being infected with BoHV-1. IBR eradication and prophylaxis measures have been assessed. As a result of that it will be possible to create new IBR eradication and prophylaxis measures to be taken in cattle farms of Lithuania. The draft of the national IBR eradication programme has been drawn up and presented. RESEARCH MATERIALS AND METHODS The study was carried out at the Department of Infectious Diseases of the Lithuanian Veterinary Academy (since 1 September 2010, the Veterinary Academy of the Lithuanian University of Health Sciences), at the Veterinary Institute of Veterinary Academy and the Virology Research Division of the Laboratory Department of the National Food and Veterinary Risk Assessment Institute between 2005 and 2011. All investigations were carried out in accordance with requirements laid down in the Order No. B1639 of the Director of the State Food and Veterinary Service of the Republic of Lithuania adopted on 22 January 2009: on keeping, breeding and using animals for experimental and other scientific purposes (Official Gazette Valstybės žinios No. 8-287, 22 January 2009). 8 Subject of investigations In 2005–2010, investigations of infectious bovine rhinotracheitis from blood serum were carried out by means of the method of the enzyme-linked immunosorbent assay. The material for serological investigations was collected from different cattle farms in Lithuania. Samples were collected from different age and sex animals at random. Cattle from 37 districts of Lithuania (n=15368) raised on randomly chosen farms and herds of different number of cattle were investigated. Samples were taken from both the farms, which had problems about health of the cattle and from the farms, which kept herds of healthy cattle and which required the possibility of BoHV-1 prevalence on these farms to be investigated. Serum samples of a total of 15368 cattle were serologically investigated. Investigations of clinical and pathological material of cattle were performed using the polymerase chain reaction. The investigations were performed in 24 herds of cattle. A total of 193 samples of clinical and pathological material were studied. Sampling methods Blood samples for serological tests were taken sterilely from the jugular or caudal vein in 10 ml vacuum tubes without anticoagulant. Blood taken was kept for one hour at room temperature, then kept at +4oC temperature overnight. Sera were poured into plastic tubes 1 ml each, numbered and kept at minus 20ºC temperature until the investigation. Lungs samples and the nasal cavity discharges for PCR tests were taken from calves aged one to twelve months. Clinical and epidemiological investigations The data about cattle productivity and the type of farm management were taken from the Department of Statistics of the Republic of Lithuania (2000 Statistical Yearbook of Lithuania, 2003 Statistical Yearbook of Lithuania). The dynamics of prevalence of cattle diseases and mortality was assessed on the basis of official yearly reports of the State Food and Veterinary Service applying the retrospective research method. For preliminary study sera samples (n=360) from randomly selected cattle in nine high productivity dairy farms were tested for antibodies to BoHV-1 at the Division of Virology Research of the National Food and Veterinary Risk Assessment Institute. To carry out comparative studies of diagnostics methods of infectious bovine rhinotracheitis three age groups of cattle were formed – cattle younger than 3 months, between the ages of 3 and 9 months, and cattle older than 9 months. Total 81 samples of bovine sera were investigated by ELISA 9 and virus neutralization (VN) methods. To compare prevalence of BoHV-1 infection with other bovine viral diseases in Lithuania, in parallel investigations into IBR, PI-3, RSV and BVD virus prevalence by ELISA were performed. For detection of antibodies to PI-3 and BVD viruses 558 samples were tested. For detection of antibodies to RSV and IBR viruses – 538 samples were tested. Seroprevalence to all mentioned viruses on 19 farms with herds larger than 200 cattle located in 11 districts has been analysed. Samples for investigations of passive and active immunity were taken from two farms, in which applied similar cattle breeding technology and kept the same number of cattle. In the farms were kept about 1500 cattle, 1000 of them were older than 3 months age. In those farms cattle were kept in very similar conditions. However, the essential difference was that the cattle of one farm were vaccinated against IBR, whereas the cattle of the other farm were unvaccinated. Seeking to establish the duration of passive and active immunity, 96 samples of blood serum were investigated. When carrying out clinical and epidemiological investigation attention was paid to the features characteristic of IBR endemics. Clinical and epidemiological investigations concerning IBR endemics were carried out in random pedigree cattle farms considering to consideration the data of serological studies of the National Food and Veterinary Risk Assessment Institute, clinical observation of the health of the herds. The material for investigations was taken from five cattle farms in which pathological anatomic and clinical symptoms characteristic of IBR were determined. During the investigation of the efficiency of infectious bovine rhinotracheitis control measures in 54 farms were analysed in which periodical IBR investigations were carried out, part in IBR eradication programmes was taken, the cattle were vaccinated, IBR monitoring was conducted and selection of the IBR seropositive cattle was made. Enzyme-linked immunosorbent assay Commercial standardised Enzyme-linked immunosorbent assay (ELISA) kits (Institute Pourquier, France and IDEXX, USA) were used for the detection of antibodies to BoHV-1 gB and gE glycoproteins and to investigate blood samples for BVD, PG-3 and RSV infections. ELISA was performed according the producer manual. Comparative evaluation of diagnostic methods In the course of investigations a comparative evaluation of standard virus neutralisation (VN) and gB ELISA methods used in IBR diagnostics was made. Blood samples collected (n=81) were divided taking into 10 consideration animal groups by age. 100 TCID50 viral particles/ 0.1 ml of BoHV-1 “4019” strain were used in the VN reaction. The data obtained by means of different research methods were compared assessing sensitivity and specificity of the reactions according to the recommendations of Kramps and the World Organization for Animal Health (Kramps et al., 1999). Polymerase chain reaction (PCR) DNR extraction from the samples of homogenate organs or discharge from the nostrils was carried out by means of phenol – chloroform- isoamyl alcohol and Genomic methods. During PCR 35 amplification cycles were completed: +95ºC 1 min., +56ºC 1 min. and +72ºC 1 min. To complete the final amplification a three-minute longer procedure +72ºC was carried out. The product of 468 nucleotide pairs was obtained during PCR. Cattle herpesvirus-1 oligonucleotides primers were used during PCR (Vilcek, 1993): P1(sense): 5´-CAC GGA CCT GGT GGA CAA GAA G-3´ and P2(antisense): 5´-CTA CCG TCA CGT GCT GTG TAC G-3´. Viruses and cell cultures The Madin-Darby bovine kidney (MDBK) cell line was used for virus propagation. Cells were cultivated in Minimum essential medium Eagle (MEM) with 10% fetal calf serum (FCS, “Biochrom”, Germany). Standard “4019” BoHV-1 strain was used in the studies. Viruses were adapted and propagated in MDBK cell cultures after 2 to 6 passages. The cell monolayer formed was washed three times with MEM without serum prior to inoculation and virus suspension was poured on the cells. The amount of viral suspension poured was such that the infecting dose should equal 0.1-1 of infecting doses of tissue cultures per cell (0.1-1 TCID50). The infected cells were incubated in an incubator at +37o C temperature for 48-72 hours up to 70-90% of the establishment of the cytopathogenic effect (CPE). Virus neutralisation reaction (VN) Prior to carrying out the VN reaction, the virus of BoHV-1 “4019” strain was titrated and its titre in MDBK cell culture amounted to 10 6,5TCID50/ml (50% of the tissue culture infection dose). The VN reaction was performed by means of micro-method using MDBK cell culture, which was not contaminated with pestiviruses, with 100 TCID50. Prior to the investigation blood serums were inactivated by heating in a water bath at +56o C temperature for 30 min. The samples were titrated on a micro-plate with 96 wells using the medium of cell cultures as a diluent. The highest dilution, in which viruses are neutralised in 50% of the wells, i.e. 11 when CPE is completely stopped, is considered to be the titre of the VN reaction. Assessment of statistical data Statistical calculations were made at the Department of Infectious Diseases of the Lithuanian Veterinary Academy of the Lithuanian University of Health Sciences, the Veterinary Institute of the Veterinary Academy and the National Food and Veterinary Risk Assessment Institute. The statistical data analysis was performed of the computer programme “Graph Prism 3.0TM”. The Student t test and the data calculated are regarded to be significant when p< 0.05. The confidence interval (CI) of serological prevalence of the BoHV-1 in a cattle population and significance of differences in the prevalence percentage, with the confidence level 95%, was calculated by means of Dimension Research, Inc. programme. RESULTS AND DISCUSSION Analysis of disease prevalence and mortality of cattle in Lithuania During the first stage of investigations, the analysis of the data of cattle treatment, which were collected until 2002 only, showed that during the period between 1999 and 2002 up to 13.9% of all cattle kept in Lithuania suffered from one or another disease. Most often diseases of digestive system and respiratory organs were recorded. In 1999, as much as 5.0% and 2.6% of cattle kept at that time, respectively, suffered from these diseases. In later years the incidence of calves falling ill with the diseases of the digestive system and respiratory organs gradually decreased, and in 2002 they accounted for 3.4% and 1.8%, respectively. When analysing the data it was established that the majority of the cattle were rejected due to udder diseases (9.7%) and reproductive disorders, infertility and genital diseases (11.9%). At that time only 1% of cattle were rejected due to infectious diseases. During the period between 1999 and 2002 of 402 thousand cattle were treated, of which 26 thousands (6.5%) of cattle died. During the period between 1999 and 2002 mainly cattle died from the diseases of respiratory organs. In 2002, the cattle mortality rate from the diseases of the digestive system increased (10.5%). However, diseases of respiratory organs were much more dangerous because many more animals died of these diseases (14%). The results of these investigations correspond with the data obtained by other investigators who maintain that 37.8% of dead calves had lung or 12 intestine injuries (Večerek et al., 2003). There are more data about the highest mortality rate of the calves from enteritis and pneumonia (Sivula et al., 1996). The larger incidence of diseases of calves and their mortality from enteritis and respiratory diseases can be accounted by a wide spread of viral diseases such as bovine viral diarrhoea, infectious bovine rhinotracheitis, rotaviral enteritis, coronaviral infection and bacterial infections (Šalomskas et al., 1997, Mockeliūnienė et al., 2004, Milius et al., 2005). Diagnostic investigations of infectious bovine rhinotracheitis in the herds of pedigree cattle During the second stage of investigations the condition of IBR was analysed in typical herds of highly productive cattle. In cattle on seven (77.8%) out of nine chosen farms were detected antibodies to BoHV-1 viruses. When analysing the data it was established that IBR was more often detected in a group of cows, especially old ones (p< 0.01). In some herds 86.7% of cows were seropositive, i.e. carriers of BoHV-1 viruses. On average, 45.5% of the cattle under investigation were infected with BoHV-1 viruses, however, it was not clear whether our chosen methods of laboratory diagnostics was sufficiently sensitive and specific, therefore during the third stage of study comparative investigations of IBR diagnostics methods was conducted. Comparative investigations of diagnostics methods of infectious bovine rhinotracheitis The comparative assessment of the Enzyme-linked immunosorbent assay of antibodies (ELISA Ab) used in the preliminary investigation of IBR serological prevalence and standard virus neutralisation (VN) methods was performed by means of each method in determining IBR seropositive animals. However, the results of the investigations into the groups of different age differed. The greatest number of inconsistencies was found in the group of calves under 3 months of age where 8.3% more seropositive individuals were found when examining them with the ELISA method than when using the standard VN method. Though these differences were not statistically significant, in practice such research results can have an influence on the quality of investigations therefore seeking to make clear differences in the results of blood serum tests received by means of the standard VN and ELISA Ab sensitivity and specificity of ELISA Ab was assessed according to the calculation schemes recommended by the World Organization for Animal Health. Though ELISA Ab was noted for high diagnostic sensitivity and specificity exceeding 90%, causes of inconsistency between the VN and ELISA Ab results were further analysed. For that purpose data of standard VN and ELISA Ab reactions were classified according to the titre groups. The highest inconsistency in ELISA Ab and VN results were obtained in the group of calves under three months of age and in the samples where antibody titres were low. Since the majority of calves of that age can have colostrial antibodies BoHV-1, this inconsistency is of no great significance to serological diagnostics of IBR infection therefore in further serological investigations namely the ELISA Ab research was applied. Scientific investigations conducted in other countries confirm that too (Van Oirschot et al., 1997, Wellenberg et al., 1998, Ballagi et al., 1999). At the present time the following serological research methods are often used to diagnose IBR infection: virus neutralisation (VN) and enzyme-linked immunosorbent assay (ELISA). These methods are most often made use of to determine antibodies to the BoHV1 virus in blood serum (Kramps et al., 1993). Besides, gE ELISA is also used in determining antibodies to the BoHV-1 virus in cattle herds in which the marker vaccine had been used (Van Oirschot et al., 1997). Epidemiological investigations of infectious bovine rhinotracheitis Having carried out the investigations provided for in the fourth stage, antibodies to infectious bovine rhinotracheitis were detected in blood sera samples of 1839 cattle, which accounted for 11.97% of all samples studied. Having made the analysis of the data of the investigations by the year of conducting the investigations, it was established that the greatest number of cattle infected with the IBR virus was in 2009 – 17.88% (CI 15.07–20.69). On the basis of the investigations performed between 2005 and 2007, a 14 tendency for the number of IBR seropositive animals to decrease was observed – in 2005 – 12.13% (CI 11.14–13.12), in 2006 – 10.80% (CI 10.04–11.56), in 2007 – 9.54% (CI 8.35–10.73). However, the investigations performed in 2008 showed that 16.68% (CI 14.93–18.43) of the cattle under investigation were IBR seropositive. In 2007, considerably fewer seropositive cattle (9.54%) than in 2009 – 17.88% (CI 15.07–20.69, p≤0.05) were found. The average of IBR seropositive cattle investigated between 2005 and 2009 accounted for 11.97% (CI 11.46–12.48). Having made the analysis of the data of the investigations by the size of cattle herds it was established that the largest per cent of IBR Ab positive cattle (14.79%) was in those cattle herds, which contained more than 200 animals. In the herds containing from one to 200 cattle the number of seropositive cattle was statistically much smaller (5.58%) than in the cattle herds containing more than 200 animals (14.79%, p≤0.05). Hence, dependence of the spread of IBR on the number of cattle in a herd was established, i.e. the more cattle were kept in one herd a larger part of cattle was infected with BoHV-1. It was also established that the greatest BoHV-1 seroprevalence was in a group of cows – 34.64% (CI 32.97–36.31), and the smallest one was among bull-calves – 2.01% (CI 1.27–2.75). BoHV-1 seroprevalence in the group of heifers accounted for 10.01% (CI 9.23–10.79), and in the group of younger animals it amounted to 4.41% (CI 3.82–5.00). The analysis of the influence of the cattle age on IBR prevalence in cattle herds was also made. Having carried out the investigations it was established that in the groups of cattle of the ages between 0 and 4 months, and between one and two years the number of seropositive cattle differed insignificantly and fluctuated from 4.26% (CI 3.7–4.84) to 6.69% (CI 5.95– 7.43). However, beginning with the age of 2–3 years the number of seropositive cattle is rapidly on the increase. In the group of animals of the ages between 2 and 3 years this percentage was 18.26% (CI 16.56–19.96), and in the group where the cattle is older than 7 years seropositive animals accounted 53.98% (CI 49.03–58.93, p≤0,05) (Fig. 1). Our epidemiological investigations showed that despite the control measures that have been taken thus far, between 2005 and 2009 from 12.13% to 17.88% of all investigated cattle was infected with BoHV-1. More often IBR was serologically diagnosed in the herds in which concentration of the cattle was great, rarer IBR occurred on small farms. Our investigations show that cows were infected with BoHV-1 most often, heifers were infected rarer, and BoHV-1 developed among bull-calves and younger animals the least. The analysis of the influence of the cattle age on IBR prevalence in cattle herds in Lithuania showed that the IBR infection 15 prevalence in the age groups of younger animals was even and depended on the age very insignificantly, however, in other cattle groups, beginning with the age of 2–3 years it had a great influence on the spread of BoHV-1 infection, i.e. the older the cattle the greater serological spread of BoHV-1 was. On the basis of our research data, the following tendency can be observed: with the number of cattle in a herd increasing, the risk of bovine BoHV-1 prevalence in a herd increases, i. e., the greater number of the cattle is kept in one herd, the more infected animals there are in it. BoHV-1 infection prevalence by cattle age groups Geographical features of infectious bovine rhinotracheitis prevalence in Lithuania The analysis of geographical BoHV-1 prevalence carried out during the fifth stage of study shows that between 2005 and 2009 the number of seropositive cattle was greatest in the following districts: in Šiauliai – 22.69%, Kaunas – 19.51%, Marijampolė – 17.26% and Utena – 15.74%. The number of seropositive cattle in other districts was much smaller: in Panevėžys – 7.77%, in Alytus – 2.76%, in Vilnius – 2.55%, in Telšiai – 1.54%, in Klaipėda – 0.85%, and in Tauragė – 0.53%. Having analysed the results of the investigations it was established that BoHV-1 infection prevailed in the districts of southern, central and northern Lithuania. 16 BoHV-1 infection investigation by means of the polymerise chain reaction (PCR) As can be seen from the investigations, BoHV-1 viruses are wide-spread in a cattle population in Lithuania. We have also established that cattle respiratory diseases, though they are in the third place according to their prevalence, are very difficult to be healed therefore animals die of them most often. Because BoHV-1 is one of the most important agents of the complex of cattle respiratory diseases, during the sixth stage of our investigations we investigated the significance of these viruses in etiology of cattle diseases. The investigations carried out by means of the PCR method confirmed that in five out of 24 cattle herds (20.83%) the cause of respiratory diseases of endemic nature of calves was BoHV-1. Having examined samples of lungs and discharge from the nostrils of 193 calves from one to twelve yearold calves, BoHV-1 nucleoacid was detected in 19 cases, or 9.84%. The data presented in the Table 2 show that BoHV-1 is usually detected in lung samples as many as 4.4 times more often than in the discharge from the nostrils of the sick calves (p=0.004). When comparing the results of the lung sample tests with the average of positive samples (9.84%) a significant difference was also obtained (p=0.037; Table 2). Table 2. The data of the investigations showed that during the outbreak of BoHV1 infection the percentage of positive samples is from 40% to 100%. Nine (52.9%) out of 17 examined lung samples were positive, when examining 16 samples of the discharge from the nostrils the number of positive ones stood at 10 (62.5%). However, this difference was not statistically significant, that is, it can be stated that these investigations were of equal value from the point of view of diagnostics. 17 The PCR method has not been applied to determine BoHV-1 viruses in the country thus far. The objective of the investigations carried out was to assess the possibilities of the polymerase chain reaction in diagnosing the acute BoHV-1 infection. Thus far it has been thought that the latent IBR form is characteristic of Lithuania, however, our PCR investigations showed that the cause of bronchopneumonia of calves could often be IBR viruses too. A rapid spread of BoHV-1 in a herd can be explained by the fact that during observation respiratory diseases were diagnosed for younger animals. The data of the investigations coincided with the data obtained by other scientists who established that BoHV-1 spread in a herd in an aerogenic way much faster than in a contact way (Mars et al., 2000). Our selected and applied the diagnostic PCR method allowed the rapid diagnosis of IBR. It was established that on all five chosen farms where calves suffered from respiratory diseases, one cause of disease and mortality was BoHV-1. The results obtained also showed that only 20.83% of cases of the outbreak of respiratory diseases were caused by BoHV-1 infection. Therefore in diagnosing respiratory diseases in calves is necessary to take into consideration the spread of other pathogens, especially parainfluenza-3 and respiratory syncytial viruses in cattle herds. Comparison of BoHV-1 infection prevalence with other bovine viral diseases in Lithuania During the seventh stage of work our investigations were aimed at making a complex analysis of the prevalence of infectious viral bovine diseases, serologically studying the prevalence of IBR, bovine parainfluenza-3 (PI-3), bovine respiratory syncytial virus infection (RSV) and bovine viral diarrhoea (BVD) in cattle herds. During the investigation were detected antibodies against all four viral diseases on most cattle farms (14 out of 19 farms, 73.7%), and only on five (26.3%) farms were detected antibodies against two or three viral diseases. When analysing the investigations carried out it was established that on 84.2% of the farms studied the cattle that had antibodies to IBR viruses were found. The cattle that had antibodies to PI-3 and RS viruses were detected on 95% of the farms studied. BVD infection was detected on 85% of farms. On average, 30.5% of cattle had antibodies to BVD, 72.2% of cattle had antibodies to PI-3, on 54.5% of cattle were detected RS viral infection, and 43.4% – BVD. The number of the seropositive cattle found differed on different farms. The number of the cattle that had antibodies to PI-3 viruses fluctuated from 28% to 93.3%. The seropositive cattle distributed themselves even more when examining RSV, IBR and BVD viral infections – from 12.5% to 100%, from 2.4% to 100% and from 6.7% to 94.4%, 18 respectively. In summing up the investigations of laboratory diagnostics of PI-3, RSV, IBR and BVD viral infections it can be stated that the said viruses are extremely widespread and might be the cause of respiratory diseases that have manifested themselves recently. Our investigations confirm the supposition that viruses causing respiratory diseases in Lithuania are very widespread, and the cause of bronchopneumonia can be not only BoHV-1 viruses causing infectious bovine rhinotracheitis. Our investigations also showed that the spread of PI-3 viruses raises especially many problems, and a similar situation is recorded in other countries of the world where intensive cattle breeding is developed (Norstrom et al., 2001, Sardi et al., 2002). It can be stated that PI-3, RS and IBR viruses, together with BVD viruses is the main cause of respiratory diseases in large herds of high productivity. Investigations into passive and active immunity During the eighth stage of work investigations into a passive and active immunity of calves were carried out. The results of blood serum tests of detection of antibodies to IBR virus glycoprotein gB is presented in Fig. 2. The part of seropositive calves in an unvaccinated group of calves (farm No. 1) changed insignificantly. The percentage dynamics of the age of seropositive calves was investigated in determining antibodies to IBR virus glycoprotein gE too. The data analysed show that all the calves up to the age of two months were seropositive in the unvaccinated herd (farm No. 1). The number of seropositive calves decreased to 12.5% in the age group of four months, whereas the number of seropositive calves in the groups of older calves constantly increased. No seropositive calves were detected in the herd of vaccinated cattle (farm No. 2) at all. When analysing the dynamics of antibodies to IBR virus glycoprotein gB it was established that antibody titres of the calves born in the herd of the unvaccinated cattle differed very insignificantly from the antibody titres of the calves born to the vaccinated cows. Note: calves of farm No. 1 were unvaccinated; calves of farm No. 2 were vaccinated on the 90th day of their life 20 The analysis of the investigations of antibodies to IBR virus glycoprotein gE in the group of the unvaccinated calves (farm No. 1) showed that the highest antibody titres (OD < 0.1) were detected for the calves in the age groups 7 and 180 days. The antibody titre in the groups of calves of the age of 120 days decreased. The lowest antibody titre of the seropositive calves was detected in the group of calves of the age of 270 days. Antibodies to IBR virus glycoprotein gE in the herd of the vaccinated calves (farm No. 2) not detected. Investigations into the dynamics of antibody titres showed that the duration of passive immunity after vaccination was shorter though antibody titres for seven-day-old calves were higher. A constant increase in antibody titres in the groups of older calves can be explained by getting infected with BoHV-1 field strain (farm No.1) and the effect of vaccination (farm No.2). The investigation into the antibodies titres to IBR virus glycoprotein gE confirmed effectiveness of vaccination. This investigation showed that infected calves were not detected in the vaccinated herd in all, whereas in the herd of the unvaccinated calves this investigation confirmed the spread of the IBR virus. Investigations into the dynamics of antibody titres carried out by other authors for the vaccinated cattle and the cattle experimentally infected with pathogenic BoHV-1 viruses also showed the efficiency of vaccine against IBR (Kaashoek et al., 1996, Bosch et al., 1998, Kamaraj et al., 2009). Investigation of effectiveness of infectious bovine rhinotracheitis control measures According to our data, by the end of 2010 year in 54 farms were applied IBR control measures. Their largest part limited themselves to periodical investigations into BoHV-1 sero-distribution (67.86%) only. In 28 farms (51.85%) carried out eradication and monitoring programmes. Ten farms (18.52%), due to a too large number of seropositive cattle, vaccinated them with marker vaccines. Hence, less than one per cent of the farms in which the control animals were kept carried out the programmes. Three of the farms had the status of the IBR free herd, which accounted for as little as 0.05% of a total number of all pedigree cattle herds. We established that only a small part of the farms applied IBR control measures. However, those 28 farms where the eradication and monitoring programmes were carried out faced with problems of implementing the programme. Hence, most often the farms failed to follow such instructions provided for in IBR eradication plans as adequate handling of documents and clinical examination of the herd performed according to the plan. Vaccination was 21 most often given adequately since both owners of the cattle and veterinary surgeons devote much attention to this part of the programme. There were eight farms (28.57%) where everything that was provided for in the programme was done. Five of them performed vaccine IBR prophylaxis. On these farms the number of the infected cattle prior to the beginning of vaccination was from 11.2% to 52.3%. Following our control investigations conducted on these farms, 20-24 months after the beginning of vaccination, it was established that after vaccination the number of the seropositive cattle decreased to 0% on four out of five farms, that is, no new BoHV-1 infection cases were diagnosed, with the exception of one farm where the number of the seropositive cattle decreased by as many as 2.4 times (from 45% to 18.6%) after vaccination. In our opinion, the vaccinated cattle do not become infected, and older seropositive cows are liquidated and their number decreases. Thus, it can be stated that constant vaccination of the entire herd is the most suitable way to eradicate BoHV-1 infection. Investigations into the efficiency of vaccination were conducted in other countries too, where it was also indicated that vaccination of cattle decreased the number of IBR cases and put a stop to the spread of BoHV-1 (Mars et al., 2001, Nardelli et al., 2008). The investigations carried out showed that on the chosen farms the majority of highly productive cows were seropositive therefore to eradicate IBR infection the method of depopulation would not be efficient in Lithuania. On the basis of experience of German, Belgian, French and Dutch scientists and veterinary surgeon-practitioners, it could be recommended to use the marker vaccine to eradicate IBR/BIPV in some herds (Eloit, 1997, Wiseman, 1997, Bosch et al., 1998, De Wit et al., 1998). The positive feature of this vaccine is that the vaccinated cattle can be distinguished from the infected ones by means of serological and molecular biological techniques (Siebert et al., 1995, Strube et al., 1996, Bosch et al., 1998). Assessment of a threat of bringing BoHV-1 to Lithuania with imported cattle IBR are widespread in Lithuania, however, it is for 5 years already that the disease is being eradicated in the 28 herds of highly productive cows, and three farms are officially free from IBR. The situation is very different in different countries of the European Union, so in buying cattle the risk of infection getting into the cattle herds of our country is different, therefore in 2004 the European Commission adopted the decision to divide the Member States of the Community in groups according to the spread of IBR viruses and to apply control measures against this disease. In 2010, the list of the 22 countries was supplemented. There is no risk of infecting our cattle with IBR in importing the cattle from Denmark, Austria, administrative units of Upper Pfalce and Upper Frankonia of Federal Land of Bavaria, Germany, the province of Bolzano in northern Italy, Finland, Sweden, a small risk remains when importing cattle from the Czech Republic, Germany – all the regions (with the exception of the administrative units of Upper Pfalce and Upper Frankonia of Federal Land of Bavaria, Germany), the autonomous region of Friuli-Veneza Giulisa in Italy and Autonomous province of Trent, a great risk of becoming infected with IBR remains when importing cattle form other EU states not mentioned here (Decision of the European Commission 2010/433/EU). According to the data of the State Animal Breeding Supervision Service under the Ministry of Agriculture, in 2006 year 959 pedigree beef cattle were imported to Lithuania. The majority of them (633 heads, or 66%) were brought from France and Ireland, that is, from the countries where IBR is widespread and the possibility of infection is high. At the same time the number of dairy heifers imported totalled 1975. The majority of them (1415 heads, or 71.6%) were bought in Germany and Denmark where IBR was either eradicated or very little widespread due to strict control. Hence, the possibility of bringing BoHV-1 viruses together with the heifers being imported is very small, with the exception of those cases when heifers were purchased in the Netherlands (21.4% of the total import of heifers in 2006). Therefore during the quarantine period the cattle are to be examined for IBR antibodies. CONCLUSIONS 1. As much as 26.7–77.8% of cattle are infected with the infectious bovine rinotracheitis virus on highly productive cattle farms, and prevalence of the IBR virus in the cattle population in Lithuania consists for 11.9%. Since 2008 an increasing tendency for a spread of viruses has been revealed. 2. Prevalence of the IBR virus depends on the size of a cattle herd. In the herds containing from one to 200 cattle the percentage of infected animals were 5.58% and in the herds with more than 200 cattle this figure stood at 14.8%. 3. The spread of the IBR virus is influenced by the age of cattle too. It was established that IBR prevalence in the group of cattle older than 7 years accounted for 57%, and the smallest prevalence was discovered in the groups of cattle of the ages between 0 and 4 months and between one and 2 years (4.3% and 6.7%). The IBR virus was extremely widespread among the 23 cows – 34.6%, and it was the least spread among the bulls – 2%. 4. ELISA Ab is noted for high sensitivity (96.3%) and specificity (90.6%) therefore it is recommended to use for serological investigations of infectious bovine rinotracheitis. 5. The PCR investigations carried out showed that only BoHV-1 infection was the cause of the outbreak of cattle respiratory diseases in 20.8% of cases therefore it is necessary to take into consideration the spread of other viruses (PI-3, RSV, BVD) in cattle herds. 6. Having made a complex analysis of the spread of viruses it was established that 30.5% of cattle had been ill with IBR, 72.2% – with PI-3, 54.5% – with RSV infection, 43.4% of cattle had been ill with BVD. It was established that on the majority of farms the cattle had been ill with four diseases, which proves that these viruses can be the cause of respiratory diseases. 7. It was established by investigations that passive immunity of the calves born to the unvaccinated cows lasted up to two months and was more durable than that of the calves born to the vaccinated cows. 8. Investigations of the efficiency of vaccination show a very high protective efficiency of vaccines on the farms where the cattle are vaccinated. No new cases of IBR infection were detected, and the number of the infected cattle decreased from 52.3 to 0%. PRACTICAL RECOMMENDATIONS To prevent IBR and other respiratory infections from getting into farms it is recommended to put the imported cattle under quarantine and to examine them by means of serological methods. In exercising IBR control on the farms it is recommended to carry out serological investigations to establish carriers of viruses. To prevent a spread of a disease on a farm, carriers of viruses and the infected animals must be separated from the healthy ones. It is necessary to ensure that the personnel servicing the farm should not spread IBR agents. It is recommended to use marker vaccines and carry out investigations according to the devised IBR eradication programmes on the farms where IBR is very widespread. P. 289–292. 25 ĮVADAS Pirmasis pranešimas apie galvijų kvėpavimo takų infekcinį rinotracheitą publikuotas 1954 metais (Schroeder and Moys, 1954). Liga buvo apibūdinta dideliu karščiavimu, agalaktija ir kvėpavimo takų pažeidimo simptomais. Vėliau buvo pasiūlytas ir priimtas ligos pavadinimas – galvijų infekcinis rinotracheitas (Mckercher et al., 1955). Galvijų infekcinio rinotracheito (GIR) sukėlėjas – Herpesviridae šeimos Alphaherpesvirinae pošeimio Varicellovirus genties virusas (BoHV-1). Tai vienas labiausiai pasaulio galvijų populiacijoje paplitusių virusų, dėl kurio daugelis galvijininkystę praktikuojančių šalių patiria didelių ekonominių nuostolių dėl sumažėjusios pieno produkcijos, apvaisinimo problemų, galvijų prieauglio netekimo, ligos sukeltų abortų. Šio viruso nešiotojai gali būti ne tik naminiai galvijai, bet ir laukiniai atrajotojai (Kendrick et al., 1987, Thiry et al., 1988, Afshar and Eaglesome, 1990, Veselinovič and Medič, 1992, Oirchot et al., 1993, Kaashoek et al., 1996, Hage et al., 1998, Frolich et al., 2002, Frolich et al., 2006). Šio viruso cirkuliacija laukinių atrajotojų populiacijoje gali turėti didelės reikšmės likviduojant ir kontroliuojant virusą naminių galvijų populiacijoje Europoje. Tuo labiau kad BoHV-1 perdavimas tarp skirtingų gyvūnų rūšių dar nėra iki galo ištirtas (Lillehaug et al., 2003, Kalman and Egyed, 2005, Frolich et al., 2006). BoHV-1 infekcija siejama su dviem pagrindiniais sindromais, vadinamais galvijų infekciniu rinotracheitu ir infekciniu pustuliniu vulvovaginitu (IPV). Šis virusas yra svarbus veiksnys, lemiantis daugelio kitų ligų, ypač galvijų respiratorinių ligų komplekso (GRLK), vystymąsi, nes BoHV-1 genomas koduoja keletą baltymų, dalyvaujančių slopinant organizmo imuninės sistemos atsaką (Turin et al., 1999, Jones and Chowdhury, 2007, Jones and Chowdhury, 2010). Nors užsikrėtus BoHV-1 virusais vystosi organizmo imuninis atsakas, virusas niekada nepasišalina iš užsikrėtusių gyvulių organizmo, ir jie visą gyvenimą išlieka latentiniais šio viruso nešiotojais (Mechor et al., 1987, Madic et al., 1995, Babiuk et al., 1996, Schynts et al., 2001, Ackermann and Engels, 2006, 188). Stresas gali sukelti latentinės infekcijos paūmėjimą, dėl to virusai gali periodiškai sklisti į aplinką. Todėl pagrindinis GIR šaltinis bandoje yra galvijai, kurie, persirgę šia liga, tampa latentiniais BoHV-1 nešiotojais ir platina virusus tiesioginio kontakto ar oro lašeliniu būdu, o buliai – su sperma (Vossen et al, 2002, Jones, 2003, Favoreel et al., 2003). BoHV-1 infekcija pasižymi didele klinikinių simptomų įvairove. Kadangi daugelio šio viruso sukeltų infekcijų eiga yra subklinikinė, turi būti taikomos kontrolės priemonės pačiai infekcijai likviduoti, o ne simptomams slopinti. GIR yra ekonomiškai labai svarbi liga. Kartu su kitomis 26 pavojingomis užkrečiamosiomis ligomis GIR kontrolė reglamentuojama Europos Sąjungos (ES) Direktyvos 64/432/EEB E priedo II dalimi. Todėl šios ligos likvidavimui rekomenduojama patvirtinti nacionalines kontrolės programas. BoHV-1 infekcija, kaip ligos priežastis, gali būti įtariama remiantis klinikiniais, patologiniais ir epidemiologiniais požymiais. Tačiau, norint patvirtinti diagnozę, reikalingi laboratoriniai tyrimai. Kompleksinė laboratorinė diagnostika šiuo atveju remiasi paties viruso arba jo komponentų ir specifinių antikūnų šiam virusui nustatymu (Parsonson and Snowdon, 1975, Mars et al., 2000). Metodai skiriasi jautrumu ir specifiškumu. Todėl yra tikslinga praktikoje taikyti skirtingus, vienas kitą papildančius metodus. Taip pat tikslinga naudotis tyrimo metodais, kuriais būtų galima tyrinėti virusų biologines savybes. Tai aktualu, vertinant GIR plitimą tarp Lietuvos galvijų ūkių. Europoje GIR iki 1996 metų buvo diagnozuojama visose šalyse, kur tik buvo atlikti tyrimai, tačiau BoHV-1 išplitimas nebuvo tolygus. Skandinavijos šalyse, kur GIR diagnozuotas rečiau, buvo pradėta ligos likvidavimo kampanija. GIR yra likviduota Danijoje, Švedijoje, Suomijoje, Austrijoje ir Italijos Bolzano provincijoje. (ES Komisijos sprendimas 2007/584/EB, Nardelli et al., 2008). Lietuvoje GIR plačiau pradėtas tyrinėti nuo 1993 metų. Tuo metu didžiausias dėmesys buvo skiriamas ligos likvidavimui bulių – spermos donorų bandose. Iki 1997 metų veislininkystės įmonėse buvo likviduoti visi seroteigiami buliai ir visos užkrėstos spermos serijos (Šalomskas et al.,1998). Vėlesni tyrimai parodė, kad įgyvendinus šias priemones veislininkystės įmonių buliai daugiau nebeužsikrėtė, nors laboratorinių tyrimų metu periodiškai buvo aptinkami pavieniai seroteigiami karantine esantys buliai (Mockeliūnas et al., 2003). Tačiau GIR padėtis pramoniniuose pienininkystės ūkiuose praktiškai netirta, anksčiau atlikti tyrimai buvo nedidelės apimties, ligos paplitimas visos šalies mastu nėra analizuotas. Ženklus GIR išplitimas Lietuvos pieninių galvijų bandose, šios ligos enzootiškumas ir atlikti tyrimai rodo, kad reikia skirti daug dėmesio šios ligos epidemiologinės situacijos nagrinėjimui, nes tik tada bus galima parengti ir efektyviai taikyti ligos diagnostikos, profilaktikos, kontrolės bei moksliškai pagrįstas ligos likvidavimo programas ir taip sumažinti GIR paplitimą galvijų ūkiuose bei šios ligos sukeliamus ekonominius nuostolius. Darbo tikslas Ištirti galvijų infekcinio rinotracheito epidemiologinę padėtį Lietuvos galvijų ūkiuose, atlikti GIR diagnostikos metodų lyginamuosius tyrimus, 27 pasyvaus ir aktyvaus imuniteto bei GIR kontrolės ir profilaktikos efektyvumo tyrimus. Darbo uždaviniai 1. Atlikti galvijų ligų paplitimo ir gaišimo priežasčių analizę Lietuvoje. 2. Nustatyti galvijų infekcinio rinotracheito viruso paplitimą Lietuvos galvijų ūkiuose. 3. Ištirti galvijų infekcinio rinotracheito epidemiologinius ypatumus. 4. Atlikti GIR laboratorinės diagnostikos metodų palyginamąjį ivertinimą. 5. Įvertinti polimerazės grandininės reakcijos galimybes diagnozuojant ūmią BoHV-1 infekciją. 6. Atlikti GIR ir kitų virusinių infekcijų (paragripo 3, respiratorinių sincitinių virusų infekcijos, galvijų virusinė diarėja virusų) paplitimo tyrimus galvijų populiacijoje. 7. Apžvelgti ir įvertinti Lietuvoje taikomas GIR prevencijos priemones ir jų efektyvumą. Darbo naujumas ir mokslinė reikšmė 1. Nustatytas GIR viruso paplitimas Lietuvos galvijų ūkiuose. 2. Nustatyta galvijų bandos dydžio, galvijų amžiaus ir lyties įtaka GIR išplitimui. 3. Nustatyta GIR virusų infekcijos dažnumo rizika pagal seroteigiamų galvijų pasiskirstymą. 4. Įvertintas diagnostikoje naudojamo antikūnų GIR virusams imunofermentinės analizės (IFA Ak) diagnostikos metodo jautrumas ir specifiškumas. 5. Įvertintos polimerazės grandininės reakcijos panaudojimo galimybės diagnozuojant ūmią BoHV-1 infekciją. 6. Palyginti GIR ir kitų virusinių infekcijų (paragripo 3, respiratorinių sincitinių virusų infekcijos, galvijų virusinės diarėjos virusų) paplitimo ypatumai galvijų populiacijoje. 7. Įvertintas GIR kontrolės ir profilaktikos priemonių efektyvumas. Darbo praktinė reikšmė Įrodyta, kad kartu taikant IFA Ak ir polimerazės grandininės reakcijos (PGR) metodus, galima užtikrinti greitą ir efektyvų GIR viruso nešiotojų nustatymą galvijų bandoje. Nustatyta, kad PGR metodas yra tinkamas aptikti BoHV-1 klinikinėje ir patologinėje medžiagoje, taip užtikrinant užsikrėtimo BoHV-1 diagnozės patvirtinimą. 28 Įvertintos GIR likvidavimo ir profilaktikos priemonės, ko pasėkoje bus galima sukurti naujas GIR likvidavimo ir profilaktikos programas galvijų ūkiams. Sukurtas ir pateiktas GIR likvidavimo programos projektas. TYRIMŲ MEDŽIAGA IR METODIKA Mokslinis tiriamasis darbas buvo atliktas 2005-2011 metais Lietuvos veterinarijos akademijoje (nuo 2010 09 01 Lietuvos sveikatos mokslų universiteto Veterinarijos Akademija) Užkrečiamųjų ligų katedroje, Veterinarijos institute ir Nacionalinio maisto ir veterinarijos rizikos vertinimo instituto Laboratorijos departamento Virusologinių tyrimų skyriuje. Visi tyrimai atlikti laikantis 2009 01 22 Lietuvos Respublikos valstybinės maisto ir veterinarijos tarnybos direktoriaus įsakymu priimto įstatymo reikalavimų: dėl gyvūnų, skirtų eksperimentiniams ir kitiems mokslo tikslams, laikymo, priežiūros ir naudojimo Nr. B1-639 („Valstybės žinios”, 2009 01 22, Nr. 8-287). Tyrimų objektas 2005–2010 metais Lietuvoje buvo atliekami galvijų infekcinio rinotracheito tyrimai iš kraujo serumo imunofermentinės analizės metodu. Medžiaga serologiniams tyrimams rinkta iš įvairių Lietuvos galvijų ūkių. Mėginiai buvo imami iš įvairaus amžiaus ir lyties galvijų atsitiktinės atrankos būdu. Ištirti 37 Lietuvos rajonuose auginami galvijai (n=15368) atsitiktinai pasirinktuose ūkiuose ir įvairaus dydžio galvijų bandose. Mėginiai buvo renkami tiek iš ūkių, kuriuose buvo problemų dėl galvijų sveikatos, tiek ir iš ūkių, laikančių sveikas bandas, kuriose buvo norima ištirti galimą BoHV-1 paplitimą. Iš viso serologiškai buvo ištirti 15368 galvijų serumo mėginiai. Galvijų klinikinės ir patologinės medžiagos tyrimai buvo atliekami naudojant polimerazės grandininę reakciją. Tyrimai atlikti 24 galvijų bandose. Iš viso ištirti 193 klinikinės ir patologinės medžiagos mėginiai. Klinikiniai ir epidemiologiniai tyrimai Tyrimams duomenys apie galvijų produktyvumą ir ūkių valdymo tipą paimti iš Lietuvos statistikos departamento (Lietuvos statistikos metraštis, 2000, Lietuvos statistikos metraštis, 2003). Galvijų ligų paplitimo ir gaišimo dinamika vertinta pagal oficialias VMVT metines ataskaitas, taikant retrospektyvųjį tyrimo metodą. GIR padėties tyrimams labai produktyvių pieninių galvijų bandose pasirinkti devyni veislinių galvijų ūkiai, kuriuose atsitiktinai parinktiems 29 galvijams (n=360) Nacionalinio maisto ir veterinarijos rizikos vertinimo instituto (NMVRVI) Virusologinių tyrimų skyriuje buvo atliekamas antikūnų BoHV-1 tyrimas kraujo serume. Galvijų infekcinio rinotracheito diagnostikos metodų lyginamiesiems tyrimams atlikti buvo sudarytos trys galvijų amžiaus grupės – jaunesni kaip 3 mėn., 3-9 mėn. ir vyresni kaip 9 mėn. Iš viso IFA ir VN metodais buvo ištirtas 81 galvijo kraujo serumo mėginys. BoHV-1 infekcijos paplitimo palyginimui su kitomis galvijų virusinėmis ligomis Lietuvoje lygiagrečiai atlikome GIR, PG-3, RSV ir GVD virusų paplitimo tyrimus imunofermentine analize. Atliekant tyrimus dėl antikūnų PG-3 ir GVD virusams, ištirti 558 mėginiai, o dėl antikūnų RSV ir GIR virusams – 538 mėginiai. Išanalizuotas šių virusų paplitimas 11 rajonų esančiuose 19 ūkių, kur buvo laikomos didesnės nei 200 galvijų bandos. Pasyvaus ir aktyvaus imuniteto tyrimams mėginiai paimti iš dviejų panašią galvijų auginimo technologiją taikančių, vienodą galvijų skaičių laikančių ir greta esančių ūkių. Siekiant nustatyti pasyvaus ir aktyvaus imuniteto laikotarpį, ištirti 96 kraujo serumo mėginiai. Klinikinių ir epidemiologinių tyrimų metu atkreiptas dėmesys į GIR endemijoms būdingus požymius. Galvijų infekcinio rinotracheito kontrolės priemonių efektyvumo tyrimo metu buvo analizuojami 54 ūkiai, kuriuose buvo atliekami periodiniai GIR tyrimai, dalyvaujama GIR likvidavimo programose, galvijai vakcinuojami, vyko GIR stebėsena ir GIR seroteigiamų galvijų atranka. Imunofermentinės analizės (IFA Ak) metodas antikūnams nustatyti Kraujo mėginių tyrimams buvo naudojami komerciniai standartizuoti imunofermentinės analizės (IFA) rinkiniai (Institute Pourquier, Prancūzija IDEX, JAV). Jie skirti antikūnams BoHV-1 virusų gB ir gE glikoproteinams nustatyti kraujo serume ir piene. Kraujo mėginių tyrimams dėl GVD, PG-3 ir RS virusų infekcijų naudoti komerciniai standartizuoti imunofermentinės analizės (IFA) rinkiniai (Institute Pourquier, Prancūzija). IFA atlikta, laikantis diagnostinių rinkinių gamintojo instrukcijų. Lyginamasis diagnostinių metodų įvertinimas Tyrimų eigoje atliktas lyginamasis standartinės virusų neutralizacijos (VN) ir gB IFA metodų, naudojamų GIR diagnostikoje, įvertinimas. Surinkti kraujo mėginiai (n=81) buvo suskirstyti, atsižvelgiant į gyvulių grupes pagal amžių. VN reakcijoje naudota BoHV-1 „4019“ padermės 100 AKID50 virusinių dalelių/ 0,1 ml, persėjamosios MDBK – galvijų embriono inkstų ląstelių kultūrų linijos. Ląstelėms kultivuoti panaudota MEM terpė (Minimum essential medium Eagle) su 10 proc. fetaliniu veršelių serumu (FVS, Biochrom, Vokietija). Duomenys, gauti skirtingais tyrimo metodais, 30 buvo lyginami, įvertinant reakcijų jautrumą ir specifiškumą pagal Kramps ir Pasaulinės gyvūnų sveikatos organizacijos rekomendacijas (Kramps et al., 1999). Polimerazės grandininė reakcija (PGR) DNR ekstrakcija iš susmulkintų organų ar šnervių ištakų mėginių buvo atliekama fenolo – chloroformo- izoamilo alkoholio ir Genomic metodais. PGR metu atlikti 35 amplifikavimo ciklai: +95º C 1 min., +56º C 1 min. ir +72º C 1 min. Galutiniam amplifikavimui užbaigti panaudota 3 min. pailginta procedūra +72º C. PGR metu gautas 468 nukleotidų porų produktas. PGR panaudoti galvijų herpesviruso-1 oligonukleotidų pradmenys (Vilcek, 1993): P1(sense): 5´-CAC GGA CCT GGT GGA CAA GAA G-3´ ir P2(antisense): 5´-CTA CCG TCA CGT GCT GTG TAC G-3´. Statistinis duomenų įvertinimas Statistiniai skaičiavimai atlikti LSMU Veterinarijos akademijos, VA Veterinarijos instituto ir NMVRVI virusologijos skyriuose. Statistinė duomenų analizė atlikta kompiuterine programa Graph Prism 3.0TM. Apskaičiuotas Stjudento patikimumo koeficientas ir duomenys laikyti patikimais, kai p< 0,05. BoHV-1 viruso serologinio paplitimo galvijų populiacijoje pasikliautinis intervalas (PI) ir paplitimo procento skirtumų patikimumas, esant 95 proc. tikimybei, apskaičiuotas Dimension Research, Inc. programa. TYRIMŲ REZULTATAI IR APTARIMAS Galvijų ligų paplitimo ir gaišimo priežasčių Lietuvoje analizė Pirmajame tyrimų etape galvijų gydymo duomenų, kurie buvo renkami tik iki 2002 metų, analizė parodė, kad Lietuvoje 1999–2002 metų laikotarpiu iki 13,9 proc. visų laikomų galvijų persirgo vienokia ar kitokia liga. Dažniausiai buvo registruojamos veršelių virškinimo ir kvėpavimo organų ligos. Šiomis ligomis 1999 metais sirgo atitinkamai 5,0 proc. ir 2,6 proc. visų tuo metu laikytų galvijų. Vėlesniais metais veršelių susirgimų virškinimo ir kvėpavimo organų ligomis palaipsniui mažėjo, ir 2002 metais buvo 3,4 proc. ir 1,8 proc. Nustatyta, kad daugiausia galvijų buvo išbrokuojama dėl tešmens ligų (9,7 proc.) ir reprodukcijos sutrikimų, nevaisingumo ir lyties organų ligų (11,9 proc.). Tuo metu dėl infekcinių ligų išbrokuojama tik 1,1proc. galvijų. Iš viso 2002 metais išbrokuota 21 296 (13,7 proc.) karvių. 1999–2002 metų laikotarpiu gydyta 402 tūkstančiai galvijų, iš jų 26 tūkstančiai (6,5 proc.) nugaišo. 1999–2002 metais daugiausia gaišdavo galvijai, sergantys kvėpavimo organų ligomis. 2002 31 metais padidėjo galvijų gaišimas nuo virškinimo organų ligų (10,5 proc.). Kvėpavimo takų ligos, kurių viena priežasčių gali būti BoHV-1 virusai, buvo daug pavojingesnės, kadangi nuo šių ligų gaišo daugiau gyvulių (14 proc.). Dėl virškinimo ir kvėpavimo organų ligų galvijai gaišo kur kas dažniau nei nuo reprodukcijos, medžiagų apykaitos ligų ar mastitų. Šie tyrimų rezultatai sutampa su duomenimis kitų tyrėjų, teigiančių, jog 37,8 proc. nugaišusių veršelių turėjo plaučių ir žarnyno pažeidimų (Večerek et al., 2003). Yra ir daugiau duomenų apie didžiausią veršelių gaišimą nuo enteritų ir pneumonijos (Sivula et al., 1996). Galvijų infekcinio rinotracheito diagnostiniai tyrimai veislinių galvijų bandose Antrajame tyrimų etape GIR padėtį analizavome tipiškose labai produktyvių galvijų bandose. Iš pasirinktų devynių ūkių septyniuose (77,8 proc.) rasta galvijų, turinčių antikūnų BoHV-1 virusams. Analizuojant duomenis nustatyta, kad GIR dažniau rastas karvių, ypač vyresnių, grupėje (p< 0,01). Kai kuriose bandose net iki 86,7 proc. karvių buvo seroteigiamos, t. y. BoHV-1 virusų nešiotojos. Vidutiniškai BoHV-1 virusais buvo apsikrėtę 45,5 proc. tirtų galvijų, tačiau nebuvo aišku, ar mūsų pasirinktas laboratorinės diagnostikos būdas yra pakankamai jautrus ir specifiškas, todėl trečiajame darbų etape atlikome GIR diagnostikos metodų lyginamuosius tyrimus. Galvijų infekcinio rinotracheito diagnostikos metodų lyginamieji tyrimai Lyginamasis antikūnų imunofermentinės analizės (IFA Ak), naudotos preliminariame GIR serologinio paplitimo tyrime, ir standartinio virusų neutralizavimo (VN) metodų įvertinimas buvo atliekamas kiekvienu metodu išaiškinant GIR seroteigiamus gyvulius. Tyrimai parodė, kad tiek VN reakcija, tiek ir IFA Ak metodais buvo nustatytas statistiškai vienodas serologiškai reagavusių galvijų skaičius (p< 0,05). Tačiau atskirų amžiaus grupių tyrimų rezultatai skyrėsi. Daugiausia neatitikimų buvo iki 3 mėn. amžiaus veršelių grupėje, kur seroteigiamų gyvulių, tiriant juos IFA metodu, buvo rasta 8,3 proc. daugiau, nei tiriant standartiniu VN metodu. Nors šie skirtumai statistiškai ir nebuvo reikšmingi, tačiau praktikoje tokie tyrimų rezultatų skirtumai gali turėti įtakos tyrimų kokybei, todėl siekdami išsiaiškinti kraujo serumų tyrimo rezultatų skirtumus, gautus standartine VN ir IFA Ak, įvertinome IFA Ak jautrumą ir specifiškumą pagal Pasaulinės gyvūnų sveikatos organizacijos rekomenduojamas apskaičiavimo schemas. Nors IFA Ak pasižymėjo dideliu diagnostiniu jautrumu ir specifiškumu, viršijančiu 90 proc., toliau 32 analizavome VN ir IFA Ak rezultatų neatitikimo priežastis. Tam tikslui standartinės VN ir IFA Ak reakcijų duomenys buvo suskirstyti pagal titrų grupes. Didžiausi IFA Ak ir VN rezultatų neatitikimai gauti iki 3 mėnesių amžiaus veršelių grupėje ir mėginiuose, kur buvo žemi antikūnų titrai. Kadangi dauguma tokio amžiaus veršelių gali turėti motininius antikūnus BoHV-1, šis neatitikimas neturi didelės reikšmės GIR infekcijos serologinei diagnostikai, todėl tolimesniuose serologiniuose tyrimuose taikėme būtent IFA Ak tyrimą. Tą patį patvirtina ir kitose šalyse atlikti moksliniai tyrimai (Van Oirschot et al., 1997, Wellenberg et al., 1998, Ballagi et al., 1999). Galvijų infekcinio rinotracheito epidemiologiniai tyrimai Atlikus ketvirtajame etape numatytus tyrimus, 1839 galvijų kraujo serumo mėginiuose buvo nustatyti galvijų infekcinio rinotracheito antikūnai, tai sudarė 11,97 proc. visų tirtų mėginių. Taip pat atlikti epidemiologiniai tyrimai parodė, kad nepaisant iki šiol taikomų kontrolės priemonių, 2005– 2009 metais nuo 12,13 proc. iki 17,88 proc. visų tirtų galvijų buvo užsikrėtę BoHV-1. Dažniau GIR serologiškai buvo nustatytas bandose, kuriose yra didelė galvijų koncentracija, rečiau GIR pasitaiko smulkiuose ūkiuose. Mūsų tyrimai rodo, kad dažniausiai BoHV-1 buvo užsikrėtusios karvės, rečiau – telyčios, o tarp buliukų ir prieauglio BoHV-1 paplitęs mažiausiai. Nustatyta, kad GIR infekcijos paplitimas galvijų prieauglio amžiaus grupėse yra tolygus ir nedaug priklauso nuo amžiaus, tačiau kitose galvijų grupėse nuo 2–3 metų amžius turi didelę įtaką BoHV-1 infekcijos išplitimui, t. y. kuo vyresni galvijai, tuo didesnis BoHV-1 serologinis paplitimas. Taip pat nustatyta, kad didėjant galvijų skaičiui bandoje proporcingai didėja galvijų BoHV-1 išplitimo bandoje rizika, t. y. kuo daugiau galvijų laikoma vienoje bandoje, tuo daugiau yra užsikrėtusių. BoHV-1 paplitimo priklausomybę nuo bandos dydžio ir galvijų amžiaus pastebėjo ir kitų šalių mokslininkai (Boelaert et al., 2000, Woodbine et al., 2009). Galvijų infekcinio rinotracheito paplitimo geografiniai ypatumai Lietuvoje 2005-2009 metais teigiamai reaguojančių galvijų daugiausiai buvo Šiaulių – 22,69 proc., Kauno – 19,51 proc., Marijampolės – 17,26 proc. ir Utenos – 15,74 proc. apskrityse. Kitose apskrityse teigiamai reguojančių galvijų buvo kur kas mažiau: Panevėžio – 7,77 proc., Alytaus – 2,76 proc., Vilniaus – 2,55 proc., Telšių – 1,54proc., Klaipėdos – 0,85 proc., o Tauragės – tik 0,53 proc. tirtų galvijų. Išanalizavus tyrimų rezultatus nustatyta, kad BoHV-1 infekcija vyrauja Pietų, Vidurio ir Šiaurės Lietuvos apskrityse. 33 BoHV-1 infekcijos tyrimas polimerazės grandinine reakcija (PGR) Tyrimai PGR metodu patvirtino, kad penkiose iš 24 galvijų bandų (20,83 proc.) endeminio pobūdžio veršelių kvėpavimo takų ligų priežastis buvo BoHV-1. Ištyrus 193 nugaišusių ar sergančių 1–12 mėnesių veršelių plaučių ir šnervių ištakų mėginius, BoHV-1 nukleorūgštis buvo nustatyta 19 atvejų, arba 9,84 proc. BoHV-1 plaučių mėginiuose rastas 4,4 kartus dažniau nei ištakose iš sergančių veršelių šnervių (p=0.004). Lyginant plaučių mėginių tyrimų rezultatus su teigiamų mėginių vidurkiu (9,84 proc.) taip pat gautas patikimas skirtumas (p=0,037). Toliau buvo analizuojama epidemiologinė padėtis bandose, kuriose kvėpavimo takų ligomis sergantiems veršeliams PGR metodu buvo diagnozuotas GIR. Tyrimų duomenys parodė, kad BoHV-1 infekcijos protrūkio metu teigiamų mėginių būna nuo 40 proc. iki 100 proc. Iš 17 tirtų plaučių mėginių teigiamų buvo 9 (52,9 proc.), tiriant 16 šnervių ištakų mėginius teigiamų buvo 10 (62,5 proc.). Tačiau šis skirtumas nebuvo statistiškai patikimas, t. y. galime teigti, kad šie tyrimai diagnostikos požiūriu buvo lygiaverčiai. BoHV-1 infekcijos paplitimo palyginimas su kitomis galvijų virusinėmis ligomis Lietuvoje Septintajame darbų etape mūsų tyrimų tikslas buvo atlikti galvijų užkrečiamųjų virusinių ligų paplitimo kompleksinę analizę, serologiškai ištirti GIR, galvijų paragripo 3 (PG-3), galvijų respiracinės sincitinės infekcijos (RS) ir galvijų virusinės diarėjos (GVD) virusų paplitimą pramoninių veislinių galvijų bandose. Nustatyta, kad daugumoje ūkių (14 iš 19, 73,7 proc.) buvo galvijų, persirgusių visomis keturiomis virusinėmis ligomis, tik penkiuose (26,3 proc.) ūkiuose buvo nustatyti galvijai, persirgę dviem arba trimis virusinėmis ligomis. Nustatyta, kad 84,2 proc. tirtų ūkių rasta galvijų, turinčių antikūnų GIR virusams. 95 proc. tirtų ūkių buvo rasta galvijų, turinčių antikūnų PG-3 ir RS virusams. GVD virusų infekcija nustatyta 85 proc. ūkių. Vidutiniškai GIR buvo persirgę 30,5 proc., PG-3 72,2 proc., RS virusų infekcija – 54,5 proc., o GVD – 43,4 proc. galvijų. Seroteigiamų galvijų įvairiuose ūkiuose rasta nevienodas skaičius. Turinčių antikūnų PG-3 virusams galvijų dalis svyravo nuo 28 proc. iki 93,3 proc. Dar labiau seroteigiami galvijai pasiskirstė tiriant RSV, GIR ir GVD virusų infekcijas – atitinkamai nuo 12,5 proc. iki 100 proc., nuo 2,4 proc. iki 100 proc. ir nuo 6,7 proc. iki 94,4 proc. Mūsų tyrimai patvirtino, kad ir Lietuvoje kvėpavimo takų ligas sukeliantys virusai yra labai išplitę, o bronchopneumonijų priežastis gali būti ne tik BoHV-1 virusai, sukeliantys galvijų infekcinį rinotracheitą. Mūsų tyrimai taip pat parodė, kad ypač daug problemų kelia PG-3 virusų 34 išplitimas, o panaši padėtis registruojama ir kitose pasaulio šalyse, kur išvystyta intensyvi galvijininkystė (Norstrom et al., 2001, Sardi et al., 2002). Galime teigti, kad PG-3, RS ir GIR virusai kartu su GVD virusais yra dažna galvijų kvėpavimo takų ligų priežastis gausiose didelio produktyvumo bandose. Pasyvaus ir aktyvaus imuniteto tyrimai Aštuntajame darbų etape buvo atliekami veršelių pasyvaus ir aktyvaus imuniteto tyrimai. Nagrinėdami duomenis matome, kad nevakcinuotų natūraliai užsikrėtusių karvių veršeliai nuo vienos savaitės iki dviejų mėnesių amžiaus visi buvo seroteigiami. Vakcinuotoje bandoje (ūkis Nr. 2) 2 mėn. amžiaus veršelių grupėje seroteigiamų veršelių sumažėjo iki 37,5 proc., o nevakcinuotų veršelių grupėje (ūkis Nr.1) seroteigiamų veršelių dalis kito nežymiai. Procentinė seroteigiamų veršelių amžiaus dinamika buvo tiriama ir nustatant antikūnus GIR viruso gE glikoproteinui. Gauti duomenys rodo, kad nevakcinuotoje bandoje (ūkis Nr.1 ) visi veršeliai iki 2 mėn. amžiaus buvo seroteigiami. Keturių mėn. amžiaus grupėje seroteigiamų veršelių sumažėjo iki 12,5 proc., o vyresnių grupėse seroteigiamų gyvulių tolydžiai daugėjo. Tuo tarpu vakcinuotų galvijų bandoje (ūkis Nr. 2) seroteigiamų veršelių iš viso nebuvo nustatyta. Analizuojant antikūnų GIR viruso gB glikoproteinams dinamiką nustatyta, kad nevakcinuotų galvijų bandoje gimusių veršelių antikūnų titrai nedaug skyrėsi nuo veršelių, gimusių iš vakcinuotų karvių, antikūnų titrų. Tačiau ūkio Nr. 1 veršelių natūralus krekeninis imunitetas buvo patvaresnis, o 60 dienų amžiaus grupėje antikūnų titrai buvo kur kas didesni nei veršelių, gimusių iš vakcinuotų karvių. Antikūnų GIR viruso gE glikoproteinui tyrimų analizė nevakcinuotų veršelių grupėje (ūkis Nr. 1) parodė, kad didžiausi antikūnų titrai (OT < 0,1) buvo nustatyti 7 ir 180 dienų amžiaus veršeliams. 120 dienų amžiaus veršelių grupėje antikūnų titras sumažėjo. Mažiausias seroteigiamų veršelių antikūnų titras nustatytas 270 dienų amžiaus veršelių grupėje. Seroteigiamų GIR viruso gE glikoproteinui veršelių vakcinuotoje bandoje (ūkis Nr. 2) nebuvo nustatyta. Antikūnų titrų dinamikos tyrimai parodė, kad povakcininio pasyvaus imuniteto trukmė buvo trumpesnė, nors antikūnų titrai 7 dienų amžiaus veršeliams buvo aukštesni. Vyresnių veršelių grupėse antikūnų titrų tolygų didėjimą galima paaiškinti užsikrėtimu BoHV-1 lauko padermėmis (ūkis Nr. 1) ir vakcinavimo (ūkis Nr. 2) poveikiu. Vakcinavimo efektyvumą patvirtino ir antikūnų titrų GIR viruso gE glikoproteinui tyrimas. Šis tyrimas parodė, kad vakcinuotoje bandoje visai nebuvo nustatyta užsikrėtusių veršelių. O 35 nevakcinuotų veršelių bandoje šis tyrimas patvirtino GIR viruso plitimą. Kitų autorių atlikti antikūnų titro dinamikos tyrimai vakcinuotiems ir eksperimentiškai patogeniniais BoHV-1 virusais užkrėstiems galvijams taip pat parodė vakcinos nuo GIR efektyvumą (Kaashoek et al., 1996, Bosch et al., 1998, Kamaraj et al., 2009). Galvijų infekcinio rinotracheito kontrolės priemonių efektyvumo tyrimas Mūsų surinktais duomenimis, iki 2010 metų pabaigos GIR kontrolės priemones taikė 54 ūkiai. Didžioji dalis jų apsiribojo tik periodiniais BoHV1 seropaplitimo tyrimais (67,86 proc.). Likvidavimo ir stebėsenos programas vykdė 28 ūkiai (51,85 proc.). 10 ūkių (18,52 proc.) dėl didelio seroteigiamų galvijų skaičiaus juos vakcinavo žymėtosiomis vakcinomis. Vadinasi, programas vykdė mažiau nei 1 proc. ūkių, kuriuose laikomi kontroliuojami veisliniai galvijai. Iš jų trys ūkiai turėjo oficialiai GIR neapimto bandos statusą, kas sudarė vos 0,05 proc. visų veislinių bandų skaičiaus. Mes nustatėmė, kad tik nedidelė ūkių dalis taiko GIR kontrolės priemones. Tačiau ir tuose 28 ūkiuose, kur buvo vykdomos likvidavimo ir stebėsenos programos, buvo programos vykdymo problemų. Dažniausiai nesilaikoma tokių GIR likvidavimo planuose numatytų veiksmų, kaip tinkamas dokumentacijos tvarkymas ir ne pagal planą atliekama klinikinė bandos apžiūra. O vakcinavimas dažniausiai atliekamas tinkamai, ir šiai programos daliai tiek galvijų savininkai, tiek ir veterinarijos gydytojai skiria daugiausia dėmesio. Ūkių, kuriuose buvo atliekami visi programose numatyti darbai, nustatėme 8 (28,57 proc.). Penkiuose iš jų vykdoma vakcininė GIR profilaktika. Šiuose ūkiuose prieš vakcinavimo pradžią užsikrėtusių galvijų skaičius buvo nuo 11,2 proc. iki 52,3 proc. Po mūsų kontrolinių tyrimų, atliktų šiuose ūkiuose praėjus 20–24 mėnesiams nuo vakcinavimo pradžios, buvo nustatyta, kad po vakcinavimo seroteigiamų galvijų skaičius keturiuose iš penkių ūkių sumažėjo iki 0 proc., t. y. nebuvo nustatyta naujų BoHV-1 infekcijos atvejų, išskyrus vieną ūkį, kuriame po vakcinavimo seroteigiamų galvijų skaičius sumažėjo 2,4 karto (nuo 45 proc. iki 18,6 proc.). Taigi mūsų tyrimai patvirtina vakcinavimo nuo GIR efektyvumą. Tyrimo rezultatai parodė, kad kai kuriose vakcinuotose bandose užsikrėtusių galvijų per tą patį laikotarpį sumažėjo nuo 52,3 proc. iki 0 proc. Mūsų nuomone, vakcinuoti galvijai neužsikrečia, o vyresnio amžiaus seroteigiamos karvės periodiškai išbrokuojamos ir jų skaičius mažėja. Taigi galime teigti, kad nuolatinis visos bandos vakcinavimas yra tinkamiausias būdas likviduoti BoHV-1 infekciją. Vakcinavimo efektyvumo tyrimai yra atlikti ir kitose šalyse, kur taip pat nurodoma, kad galvijų 36 vakcinavimas sumažina GIR sergamumą ir sustabdo BoHV-1 plitimą (Mars et al., 2001, Nardelli et al., 2008). BoHV-1 patekimo į Lietuvą su importuojamais galvijais grėsmės įvertinimas Lietuvoje GIR sukėlėjai yra labai išplitę, tačiau jau penktus metus 28 didelio produktyvumo pieninių karvių bandose liga likviduojama, o trys ūkiai yra oficialiai laisvi nuo GIR. Įvairiose ES šalyse padėtis yra labai skirtinga, tad perkant galvijus skirtinga ir rizika užkratui patekti į mūsų šalies galvijų bandas. 2006 metais į Lietuvą buvo importuoti 959 veisliniai mėsiniai galvijai. Dauguma jų (633 vnt., arba 66 proc.) buvo atvežti iš Prancūzijos ir Airijos, t. y. šalių, kur GIR yra išplitęs ir užsikrėtimo galimybė didelė. Tuo pat metu veislinių pieninių telyčių įvežta 1975. Dauguma jų (1415 vnt., arba 71,6 proc.) pirktos Vokietijoje ir Danijoje, kur GIR arba likviduota, arba labai mažai išplitusi dėl griežtos kontrolės. Taigi BoHV-1 virusų patekimas su įvežtomis telyčiomis yra mažai tikėtinas, išskyrus tuos atvejus, kai telyčios buvo pirktos Nyderlanduose (21,4 proc. viso telyčių importo 2006 m.). Todėl karantinavimo metu galvijus reikia tirti dėl GIR antikūnų. IŠVADOS 1. Labai produktyvių galvijų ūkiuose 26,7–77,8 proc. galvijų yra užsikrėtę galvijų infekcinio rinotracheito virusu, o GIR viruso paplitimas Lietuvos galvijų populiacijoje sudaro 11,9 proc., nuo 2008 metų nustatyta didėjanti BoHV-1 plitimo tendencija. 2. GIR viruso paplitimas priklauso nuo galvijų bandos dydžio. Bandose nuo 1 iki 200 galvijų užsikrėtusių individų buvo 5,58 proc., o bandose, kuriose laikoma daugiau kaip 200 galvijų – 14,8 proc. 3. GIR viruso plitimui įtakos turi galvijų amžius. Nustatyta, kad vyresnių kaip 7 metų amžiaus galvijų grupėje GIR paplitimas – 57 proc., o mažiausias– 0–4 mėn. ir 1–2 metų galvijų grupėse (4,3 proc. ir 6,7 proc.). Labiausiai GIR virusas paplitęs yra tarp karvių– 34,6 proc., mažiausiai – tarp bulių– 2 proc. 4. IFA Ak pasižymi dideliu jautrumu (96,3 proc.) ir specifiškumu (90,6 proc.), todėl rekomenduotina ją naudoti galvijų infekcinio rinotracheito serologiniams tyrimams. 37 5. Atlikti PGR tyrimai parodė, kad tik 20,8 proc. atvejų galvijų kvėpavimo ligų protrūkių priežastis buvo BoHV-1 infekcija, todėl būtina atsižvelgti ir kitų virusų (PG3, RSV, GVD) išplitimą galvijų bandose. 6. Atlikus kompleksinę galvijų virusų paplitimo analizę nustatyta, kad GIR persirgę – 30,5 proc., PG3 – 72,2 proc., RSV infekcija – 54,5 proc., GVD – 43,4 proc. galvijų. Daugumoje ūkių nustatyta, kad galvijai yra persirgę visomis keturiomis ligomis, ir tai įrodo, kad šie virusai gali būti kvėpavimo ligų priežastis. 7. Nustatyta, kad veršelių, gimusių iš nevakcinuotų karvių, pasyvus imunitetas trunka iki 2 mėn. ir yra patvaresnis nei veršelių, gimusių iš vakcinuotų karvių. 8. Vakcinavimo efektyvumo tyrimai rodo labai didelį vakcinų apsauginį efektyvumą – ūkiuose, kuriuose atliekamas vakcinavimas , nebuvo nustatyta naujų GIR atvejų, o užsikrėtusių galvijų sumažėjo nuo 52,3 iki 0 proc PRAKTINĖS REKOMENDACIJOS GIR ir kitų respiratorinių infekcijų patekimui į ūkius keliui užkirsti rekomenduojama atvežtus naujus galvijus karantinuoti ir juos ištirti serologiniais metodais. Atliekant GIR kontrolę ūkiuose rekomenduojama atlikti serologinius tyrimus, nustatant virusų nešiotojus. Užkertant kelią ligos plitimui ūkyje nustatyti virusų nešiotojai ir sergantys galvijai turi būti atskirti nuo sveikų. Užtikrinti, kad ūkį aptarnaujantis personalas neplatintų GIR sukėlėjų. Rekomenduojama ūkiuose, kuriuose GIR yra labai išplitęs, naudoti žymėtas vakcinas ir nuolat atlikti tyrimus pagal sudarytas GIR likvidavimo programas.
