Product Description
Anti-Rat Ciliary Neurotrophic Factor was developed in
goat using recombinant, rat ciliary neurotrophic factor
(CNTF), expressed in E. coli, as the immunogen. The
antibody is purified by Protein G affinity chromatography. Goat Anti-Rat CNTF is provided lyophilized
from phosphate buffered saline, to which no
preservatives have been added.
CNTF was initially identified as a trophic factor for
embryonic chick ciliary parasympathetic neurons. CNTF
is also a survival factor for developing ciliary ganglionic
neurons.1 Rat CNTF was initially purified from rat
sciatic nerve extract.1 The rat CNTF coding domain
consists of one intron. 1 The amino acid sequence of rat
CNTF has 85% homology to human and rabbit CNTF.1
Ciliary Neurotrophic Factor (CNTF) is produced in E.
coli by a DNA sequence encoding the rat CNTF
protein. 2
Performance
Anti-Rat CNTF is tested for its ability to neutralize the
biological activity of recombinant rat CNTF activity on
the human cell line, TF-1. The ND50 of the antibody is
defined as the concentration of antibody resulting in a
one-half maximal inhibition of bioactivity of rat,
recombinant CNTF that is present at a concentration
just high enough to elicit a maximum response.

Bioactivity:
ND50 = 50-100 µg/ml
Indirect ELISA: 1 µg/ml detects 0.2 ng/well of recombinant, rat CNTF.

Immunoblotting: 2 µg/ml antibody detects recombinant,
rat CNTF at 2 ng/lane under both
reducing and non-reducing conditions
Sterility:
0.2 µm-filtered, aseptic fill
Reconstitution and Use
To one vial of lyophilized powder, add 1 ml of sterilefiltered PBS to produce a 1.0 mg/ml stock solution of
Anti-Rat CNTF. If aseptic technique is used, no further
filtration should be needed for use in cell culture
environments.
Storage
Prior to reconstitution, store at -20ºC. Reconstituted
product may be stored at 2-8ºC for a maximum of one
month. For prolonged storage, freeze in working aliquots
at -20ºC. Avoid repeated freezing and thawing.
