Investigating particle features

analysis services. The image analysis is objective, fully repeatable and uncovers particle features

Particle assessment services
By analysing several aspects of VPs and VLPs, Vironova provides customers with deep insight into their particles’ quality and characteristics in
the final product or after individual purification or production steps.
A standard particle assessment package includes the following studies of the material:
Size distribution
Most processes for expressing VLPs in cell cultures generate particles
of different sizes. Generally many particles are outside the desired size
range, which impairs the desired properties of the end product. This
service provides statistically reliable data on the number of particles and
their size distribution. For example the service can be used to evaluate
different expression protocols to optimise production.
Morphology assessment
This service provides customers with information on the homogeneity
of a sample or batch, and whether there are different particle populations present. This is of great importance to optimise features and to
ensure the desired effect of particles.
Packaging
Packing of VPs/VLPs for drug or gene delivery is never 100% successful.
TEM is an excellent tool to observe the particles as a population and to
measure the full-to-empty particle ratios. This is often done manually by
ocular counting, a cumbersome and subjective process. Vironova’s service provides customers with objective data on the degree of packaging for each particle and on the distribution of packed and unpacked
particles in the batch. This is valuable information for evaluating the
efficacy of the production protocol.
Aggregation
Aggregation inhibits the efficacy of particle based vaccines or viral
vectors. This service gives valuable insight into the degree of aggregation in all steps of production. Particle aggregation is often influenced by the physical conditions of the medium. With this service the
optimal buffer conditions can be determined to avoid aggregation.
Broken particles and/or cell debris
Purification of VPs/VLPs is a trade-off between preserving as many particles as possible and having as little debris as possible. This service helps
our customers monitor the amount of broken particles and cell debris
in relation to intact particles.
