Ευχαριστώ. racellular region of EGFR consists of four domains, the ligand-binding domains (L1 and L2) an teine-rich domains (CR1 and CR2), and the C-terminal domain of EGFR contains six tyrosine r . Ras–Raf–MEK–MAPK pathway is activated through the growth factor receptor-bound protein RB2)–SOS complex. e phosphatidylinositol 3-kinase (PI3K)–AKT pathway, which contributes to optotic effects of EGFR activation. signal transducer and activator of transcription (Stat) prote TAT1, STAT3 and STAT5) are also activated. The coordinated effects of signalling pathways lead uction of cellular responses including proliferation, differentiation, cell motility, adhesion and giogenesis. e deregulation of EGFR-mediated signalling in some cancer cells leads to aberrant proliferation asion, metastasis and neovascularization. all-molecule tyrosine kinase inhibitors (TKIs) function as ATP analogues and inhibit EGFR nalling by competing with ATP binding within the catalytic kinase domain of RTKs. Each a different selectivity for RTKs, and some are dual- or multi-selective erapeutic monoclonal antibodies (mAbs) bind to the ectodomain of the RTK with high specific like small-molecule inhibitors, mAbs also activate Fcγ-receptor-dependent phagocytos olysis by immune-effector cells such as neutrophils, macrophages and natural killer cells ucing complement-dependent cytotoxicity (CDC) or antibody-dependent cellular cytotoxicity gand (L) binding to the extracellular domain of the treceptor causes dimerization that esults in autophosphorylation and activation of the intracellular kinase domain (K). hosphorylation of docking sites on the receptor leads to the recruitment of enzymes. gnalling pathways regulate transcription, translation, metabolism, cell proliferation, urvival, differentiation and motility. Gene mutations or overexpression can trigger A tyrosine kinase is an enzyme that can transfer a phosphate group from ATP to a protein in a cell. It functions as an "on" or "off" switch in many cellular functions. Tyrosine kinases are a subclass of protein kinase. The phosphate group is attached to the amino acid tyrosine on the protein. Phosphorylation of proteins by kinases is an important mechanism in communicating signals within a cell (signal transduction) and regulating cellular activity, such as cell division. Protein kinases can become mutated, stuck in the "on" position, and cause unregulated growth of the cell, which is a necessary step for the development of cancer. Most tyrosine kinases have an associated protein tyrosine phosphatase, which removes the phosphate group. Protein kinases are a group of enzymes that possess a catalytic subunit that transfers the gamma (terminal) phosphate from nucleotide triphosphates (often ATP) to one or more amino acid residues in a protein substrate side-chain, resulting in a conformational change affecting protein function. The enzymes fall into two broad classes, characterised with respect to substrate specificity: serine/threoninespecific and tyrosine-specific The mechanistic target of rapamycin (serine/threonine kinase) (MTOR; also known as mammalian target of rapamycin, mTOR. a serine/threonine protein kinase . belongs to the PI3K-related kinase protein family. The discovery of mTOR was made a few decades ago while investigating the mechanism of action of its inhibitor, rapamycin. It is a macrolide Rapamycin was discovered in a soil sample from Easter Island, known locally as Rapa Nui, in the 1970s. The bacterium Streptomyces hygroscopicus, isolated from that sample, produces an antifungal that researchers named rapamycin after the island. Rapamycin arrests fungal activity at the G1 phase. it suppresses the immune system by blocking the G1 to S phase transition in T-lymphocytes. Thus, it is used as an immunosuppressant following organ transplantation Everolimus (RAD-001) as an inhibitor of mammalian target of rapamycin (mTOR). used as an immunosuppressant to prevent rejection of organ transplants and treatment of tumours.. Progressive or metastatic pancreatic neuroendocrine tumors not surgically removable (May 2011) its effect is solely on the mTORC1 protein complex and not on the mTORC2 complex. This can lead to a hyper-activation of the kinase AKT via inhibition on the mTORC1 negative feedback loop while not inhibiting the mTORC2 positive feedback to AKT. This AKT elevation can lead to longer survival in some cell types. Hence, Everolimus plays an important role in cell growth, cell proliferation and cell survival. TSC1 and TSC2 (which are the genes involved in tuberous sclerosis disease) act as tumor supressor genes by regulating mTORC1 activity. Thus, either the loss or inactivation of one of these genes lead to the activation of mTORC1. Everolimus binds to its protein receptor FKBP12, which directly interacts with mTORC1 inhibiting its downstream signaling. As a consequence, mRNAs that codify proteins implicated in the cell cycle and in the glycolisis process are impared or altered, so tumor growth is inhibited. Hence, Everolimus inhibits tumor cells' growth and proliferation.
