SOB and SOC media (for bacteria) For SOB Media (1L) Add 20g Bacto Tryptone Add 5g Bacto Yeast Extract Add 2ml of 5M NaCl Add 2.5ml of 1M KCl Add 10ml of 1M MgCl2 Add 10ml of 1M MgSO4 Adjust to 1L with distilled H2O Sterilize by autoclaving (45 minutes, liquid cycle) (for plates): Add enough Bacto-Agar to result in a 1.5% agar solution (e.g., 15 g/1L) and autoclave. ------------------------------For SOC media (100 mL) 99 mL of SOB media + 1 mL of 2M glucose solution [Final glucose concentration of 20 mM] [Note, can become contaminated easily!]
