Résumé du projet de thèse (1 page maximum, en anglais) Translation initiation accounts for a major part of gene expression regulation, and its perturbations has deleterious effect, notably in cancer. Besides the general, cap-dependant, translation initiation process where an initiation complex scans the 5’UTR of mRNA from the 5’cap to find the initiation codon, an alternative initiation process exists. The initiation complex binds closer to the initiation codon thanks to an Internal Ribosome Entry Site (IRES). Such system is widely used by viruses, where such RNA structures where shown to function by capturing the preinitiation complex or mimick initiation factors (eIFs). Cellular IRES of non-viral origin account for about 10% of protein production in human, mostly, interestingly, when the eIFs are less active, in conditions of apoptosis, amino acid starvation, hypoxia or mitosis. In cellular IRES, the structure of the RNA is more elusive, and requires a protein (ITAF, IRES-trans acting factor) to function. An interesting model to study translational control is sea urchin, where the first cellular cycles of early development are supported by maternal messengers RNAs, and fertilization triggers an activation of the translation machinery. A global analysis realized in the team showed that among the RNAs translated following fertilization, some produce proteins identified previously as possible ITAF, such as DAP5. Protein DAP5 has been found in human to function as a platform for assembly of initiation factors, including eIF3, which directly binds the 40S small subunit of the initiating ribosome. Human DAP5 has been shown to bind RNAs, but what determines and what results of this event is still to be discovered. The project is to study the RNA-binding function of DAP5 in sea urchin with a structural approach. Search for the IRES motif will be made with a bioinformatics analysis of the RNAs translated in the first cycles and biochemically in urchin cell extracts (RNA-immunoprecipitation). The protein will be prepared as recombinant to monitor direct binding with RNA candidates. Complexes will be prepared for crystallization and analyzed by Small Angle X-rays Scattering. We expect to unveil the mysteries of cellular IRESs. Candidates should be highly motivated.
