Electrical Safety
The following items should be checked prior to initiating
any experiment in electrophoresis.
Important: The instrument must always be used with
the earth lead of the power cord correctly grounded to
earth at the mains outlet.
Work area. The bench and floor should be dry.
High-voltage connections. The high-voltage leads
should be intact and not frayed. The plug should have a
protective plastic sleeve that shields the plug as it is inserted into the power supply. Exposed plugs are a shock hazard and should be replaced with shielded plugs. Stackable
leads that connect more than one gel unit to a single outlet are not recommended and should be replaced with
shielded-style plugs.
Electrophoresis chambers. These should be covered
when in use, with no openings large enough to allow fingers or other objects to make contact with the electrified
buffer or electrodes.
Power supply. The SX 250 power supply has recessed
outputs that minimize the possibility of contacting an
electrically active plug or high-voltage input jacks.
In case of malfunction, do not open the instrument. Please
contact Pharmacia Biotech technical service.

Securité Electrique
Les points suivant doivent être vérifiés avant de commencer toute experience électrophorétique.
Important: l’instrument doit toujours être utilisé propremment connecté à la prise de terre.
Place de travail: la paillasse et le sol doivent être secs.
Connections de haut voltage: les prises de courant à
haut voltage doivent être intactes et non effilochées. Les
prises ont une manche protectrice en plastique qui les
protègent quand elles sont branchées au générateur de
courant. Des prises exposées (non protégées) sont un
danger d’électrocution et doivent être remplacées par
des prises avec protection. L’utilisation de multi-prises qui
permettent de brancher plus d’un gel sur une sortie électrique du générateur ne sont pas recommandées et
devraient être remplacées par des prises protégées.
Cuves électrophorétiques: elles devraient être couvertes durant leur utilisation. Assurez-vous qu’il n’y aie
aucune ouverture suffisement grande pour permettre à
des doigts ou autre objets d’occasionner des contacts
avec le tampon ou les électrodes.
Le generateur: le SX 250 a des sorties électriques très
enfoncées ce qui minimisent les risques de contact avec
une prise active ou une prise de haut-voltage.
Ne pas ovrir l’appareil en cas de mauvais fonctionnement.

Power Supply Function
The SX 250 power supply is intended for small-format
electrophoresis applications. It can be operated at
constant voltage (1 to 250 VDC @ 1 to 200 mA) or
constant current (1 to 200 mA @ 1 to 250 VDC) with
better than 1% accuracy.
Voltage is selectable in 1 V increments, and current in
1 mA increments. The setting or operating value is
displayed on a 3-digit LED meter. There are two pairs
of recessed 4 mm output jacks.
SX 250 Hook Up
1 Connect the mains power cord to the power cord
receptacle on the rear panel and to a suitable
grounded 3-wire AC power outlet. Place the power
supply so that the back and bottom vents are not
obstructed.
2 Connect the gel box leads to the power supply by
plugging the connectors into the recessed output
jacks.
3 Turn on the mains power switch on the rear panel.
The display will read OFF when the unit is ready.

The following power connection protocol
should minimize hazards and is recommended for all power supplies:
1 Start with the power supply off and the voltage and
current controls set at zero.
2 Connect the gel box and leads to the power supply.
Caution
When connecting high-voltage leads to the power supply, use your right hand only. Because of the potential for
lethal shock across the chest do not use both hands to
plug in (or unplug!) power supply leads. Also, make sure
your left hand is not touching anything that would
ground you.
3 Turn on the power supply and set for the desired current or voltage.
4 At the end of the run, set the voltage and current to
zero and then turn off the power supply at the AC
mains.
Caution
Power supplies have internal capacitance that stores electrical charge even after the power supply is turned off.
This stored charge can deliver a potentially lethal shock
should the operator come in contact with the positive
and negative output. Bringing the voltage and current
display to zero indicates the power supply can be safely
turned off.
5 Unplug the high-voltage leads using your right hand
only.

Set Run Parameters
1 To set parameters, press SET/READ to select the set
mode. A green lamp above the SET/READ button indicates the set mode is selected.
2 Press METER READ to select the parameter. A green
lamp lights next to either VOLTS or mAMPS. (Press
again to switch to the other parameter.)
3 Press ADJUST + or – to adjust the display value, which
increases by increments of 1 for the first 10 counts,
and then by increments of 10.
4 Repeat steps 2 and 3 for the other parameter if
required.
Note: Ten seconds after setting a parameter, the display automatically switches to READ mode. If the DC
output is off, OFF will display. If the DC output is on,
the voltage will display.

Start the Power Supply
1 Press START/STOP to activate DC output. The VOLTAGE
ON lamp lights, and the parameters last set are displayed.
Note: The default start mode is read voltage.
2 Press METER READ to display output current or voltage. A green lamp lights next to the parameter displayed.
3 Press START/STOP to turn off the DC output.

Troubleshooting
Display shows –E–
✓ Check that neither the voltage or current setting
is zero. Press any key to reset and then set nonzero parameters.
Open circuit lamp lit or display shows –OC
✓ Check that both leads from each electrophoresis
unit are plugged in completely.
✓ Check that both electrophoresis unit electrodes
contact buffer and that buffer contacts both
ends of the gel.
✓ Check for broken leads or electrodes.
No display
✓ Check that the power supply is plugged into a
working receptacle.
✓ Check input fuses.

Power Supply Crossover
A lamp next to the display indicates the parameter currently held constant. “Crossover” occurs when the second
parameter limit is reached as system resistance changes.
At this point the new parameter is held constant (the
other lamp lights) and the previously unchanging parameter is allowed to adjust downward as system resistance
continues to change. (System resistance is affected by discontinuous buffers and changes in temperature.)

Care and Maintenance
Cleaning
Warning!
If a spill leaks onto the circuit boards do not
plug the unit into a power outlet. Call the
Pharmacia Biotech Technical Service
Department for advice before using.
Caution!
Turn the mains power switch off and unplug
the power cord before cleaning.
Use a soft damp cloth dampened with water or a mild
cleaning solution to clean the cabinet and display.
Replacing fuses
Caution!
Turn the mains power switch off and unplug
the power cord before changing fuses.
Important! For continued protection against fire,
replace only with the same fuse type and
rating: 115 V model—0.8 A slow blo, 5x20,
230 V model—0.4 A slow blo, 5x20.
Two fuse holders are accessible at the rear panel. Each
fuse is held in a spring-loaded shell that partially slides
out. To open, rotate ¼ turn to the left until the cap pops
outward. Allow the fuse to slide out. Place a new fuse in
the holder and secure the shell.
