Collaboration between laboratory of molecular microbiology of human pathogens unit,
Instituto de tecnología química e biologíca (ITQB), Universidades nova de Lisboa (UNL),
Oeiras, Portugal and Pediatric infectious diseases research group (PIDRG-HSJD), Hospital
Sant Joan de Déu, University of Barcelona.
Dr. Raquel Sá-Leão from the Laboratory of Molecular Microbiology of ITQB and Prof.
Hermínia de Lencastre head of the Molecular Microbiology of Human Pathogens Unit are
worldwide recognized authorities in the field of pneumococcal infections. They have
important experience about genetic studies related to the diversity of pneumococcal
populations.
PIDRG-HSJD is an emergent research group in the field of pneumococci in Spain. This group
has a well characterized collection of invasive and carriage pneumococcal isolates among
children from Catalonia, during the period 2004-2009. In addition, the molecular
microbiology department of HSJD has wide experience in the use of molecular microbiology
techniques such as Real-Time PCR and sequence-based methods for the diagnosis and
characterization of main pediatric infections. In the field of pneumococci, Real-Time PCR is
applied for diagnosis of pneumococci in direct biological samples (plasma, cerebrospinal
fluid or pleural fluid among others) molecular capsular type detection of pneumococci and
pneumococcal bacterial load quantification. Automatic sequencing is applied for clonal type
detection of pneumococci by MLST.
The objective of this collaboration is to exchange information and know-how through handon experiments about:
1) Real-Time PCR applications for pneumococcal detection and molecular serotyping
2) Serotyping methods based on bacterial culture
3) Detection and characterization of atypical pneumococci
4) Application of Simpson’s diversity index and other coefficients to compare pneumococcal
collections
To develop this collaboration, Dra. Carmen Muñoz-Almagro, Head of PIDRG-HSJD and
responsible of the Molecular Microbiology Department of HSJD, will make a formative
internship next July 2011 in ITQB in Oeiras, Portugal.
This stage will serve to complete the study “ Invasiveness of pneumococcal serotypes and
clones circulating in Catalonia during heptavalent conjugate vaccine period” performed by
Carmen Muñoz-Almagro and the PIDRG-HSJD under the supervision and collaboration of Dr
Raquel Sá-Leão and Prof. Hermínia de Lencastre.

The study compares the incidence of serotype 19A before and after the introduction of conjugate
pneumocccal vaccine. The conclusion of the study reported that in our geographical area there was
an increase of IPD caused by S. pneumoniae serotype 19A which was mainly related with the
emergence of preexisting clones several of them closely related with international multidrug-resistant
clones.
