Carcinogenesis:
The Molecular Basis of Cancer
Four fundamental principles of carcinogenesis: {very well-illustrated in the next figure, a
flow chart showing a simplified scheme of the molecular basis of cancer}.
(I) Nonlethal (non-killing) genetic damage (or mutation) lies at the
heart of carcinogenesis. Such mutation may be:
Inherited in the germ line, or it may be Acquired by the action of
environmental agents, such as chemicals, radiation, or viruses.
(II) Three classes of normal regulatory genes;
(1) growth-promoting protooncogenes;
(2) growth-inhibiting cancer suppressor genes,
(3) genes that regulate apoptosis, are the main targets of genetic damage.
