Product Description
Monoclonal Anti-Tis7 (mouse IgG2a isotype) is derived
from the hybridoma IRT537 produced by the fusion of
mouse myeloma cells (NS1 cells) and splenocytes from
Balb/c mice immunized with a synthetic peptide
corresponding to amino-acids 30-47 of mouse Tis7,
conjugated to KLH. The isotype is determined using a
double diffusion immunoassay using Mouse Monoclonal
Antibody Isotyping Reagents (Sigma ISO-2).
Monoclonal Anti-Tis7 recognizes human and mouse Tis7
(~50 kDa). The antibody may be used in ELISA,
immunoblotting, and immunocytochemistry.
Tis7 [also known as TPA-Induced Sequence 7 (in mice),
PC4 (in rat) and Interferon-Related Developmental
Regulator 1 (IFRD1 in humans)] was cloned by differential screening for cDNAs that are up regulated during
nerve growth factor (NGF) induced neural differentiation

in rat PC12 cells. In adult rats, Tis7 is expressed in
skeletal muscle, heart and spleen but not in kidney, liver
and brain. In the embryo, the gene is expressed in the
brain, placenta and amnion at 13.5 days of development. Human Tis7 (IFRD1) protein is 90% identical to
both rat and mouse proteins, and 48% identical to
human SKMc15. Tis7 gene is up regulated upon Juninduced loss of polarity in a mouse mammary epithelial
cell line. In the un-polarized cells, the Tis7 protein
translocates into the nucleus. This protein was found to
interact with several proteins of the SIN3 complex,
including Sin3b, HDAC1, Ncor, and Sap30. This
interaction is important for the repression of transcription

of other genes. Tis7 knockout mice were fertile and
showed no gross histologic abnormalities. However,
there was delayed skeletal muscle regeneration and
altered skeletal muscle isometric contractile properties
after muscle crush damage and down regulation of
expression of several muscle-specific genes. Cultured
primary myogenic satellite cells from Tis7-/- mice
showed reduced differentiation potential and fusion

index.
Reagent
The antibody is supplied as a solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium
azide.

Storage/Stability
For continuous use, store at 2-8 °C for up to one month.
For extended storage, freeze in working aliquots.
Repeated freezing and thawing is not recommended.
Storage in “frost-free” freezers is also not recommended. If slight turbidity occurs upon prolonged
storage, clarify the solution by centrifugation before
use. Working dilution samples should be discarded if
not used within 12 hours.
Product Profile
By immunoblotting, a working antibody concentration of
0.25-0.5 µg/mL is recommended using extracts of
COS7 cells transfected with mouse Tis7.
Note: In order to obtain the best results using various
techniques and preparations, we recommend determining optimal working dilutions by titration.
