Rapid OneStep Tissue/Swab Extraction protocol Introduction Rapid OneStep Tissue/Swab Extraction protocol is intended for use with fresh tissue
or swab samples and yields appropriate amounts of DNA for successful amplification
of targets via PCR and qPCR methods.
Protocol 1. Add 100µl of Rapid OneStep Tissue/Swab Extraction buffer I for every 3x3
mm fresh tissue sample or one swab pre-seprated from stick by hand or
scissors. Make sure the pieces of swab are submerged underneath the
extraction buffer
2. Incubate samples at 95°c for 10 minutes.
3. Centrifuge sample at > 10,000 rpm for one minute and transfer supernatant to
a fresh tube.
4. Add approximately 30% of Rapid OneStep Tissue/Swab Extraction buffer II
to the sample and briefly vortex mixture.
5. Samples can be kept for several days at 4°c or for several months at -20°c.
Storage Rapid OneStep Tissue/Swab Extraction buffer should be kept at Room Temperature.

Protocol 1. Cur the lower portion (root) of one to three hair/freshly plucked samples (5
mm)
2. Add 100µl of Rapid OneStep Tissue/Swab Extraction buffer I and make
sure the pieces are submerged underneath the extraction buffer
3. Incubate samples at 95°c for 10 minutes.
4. Centrifuge sample at > 10,000 rpm for one minute and transfer supernatant to
a fresh tube.
5. Add approximately 30% of Rapid OneStep Tissue/Swab Extraction buffer II
to the sample and briefly vortex mixture.
