Haploid cellular models represent an attractive alternative
for in vitro genetic analyses of molecular, cellular, and
developmental events. Haploid cellular models are easier to
generate than modified diploid cell lines, as only one allele
needs to be targeted to obtain a complete loss-of-function
phenotype.
The HAP1 cell line is a near-haploid human cell line that
allows rapid gene editing to be performed. HAP1 cells are
adherent with fibroblast-like morphology (Figure 1). These
cells are ideal for applications where a workhorse cell line
is needed for rapid hypothesis testing, similar to the way
HEK 293 and HeLa cells have been used to accelerate
research programs for years. The GeneArt Engineered Cell
Models collection includes knockout cell lines generated
using HAP1 cells and a wide range of other cell lines.

HAP1 is the workhorse cell line for genome
editing experiments
Knockout cell lines are the most powerful in vitro models
available for understanding gene function. One of the
challenges to the rapid generation of knockout cell lines in
mammalian cell models is the diploid nature of the genome,
which requires mutations that eliminate gene function
to be introduced into both copies of the gene to ensure
complete knockout.
