Aldose Reductase is purified from the culture medium containing baculovirus-insect cell line 32d gene expression system. cDNA containing the entire protein coding region is expressed, and the enzyme exhibits the same properties and characteristics of human muscle and retina. Aldose Reductase catalyzes the reduction of glucose to sorbitol. Sorbitol is subsequently converted to fructose by sorbitol dehydrogenase. These two enzymes are key components of the polyol pathway, the alternate route of glucose metabolism. It is suggested that aldose reductase is involved in the pathogenesis of diabetic complications and the inhibitors continue to be developed at a vigorous pace.
