Stage proposé par Jean Pierre SIFFROI Professeur des Universités et Praticien-Hospitalier Nom et adresse du Laboratoire : Service de Génétique et d’Embryologie médicales Hôpital Armand Trousseau Téléphone : 01 44 73 52 80 ou 01 44 73 52 82 Mail : [email protected] Directeur du Laboratoire : Pr Jean Pierre SIFFROI Titre : Fragmentation de l’ADN des spermatozoïdes chez les sujets porteurs de translocations chromosomiques Projet de stage : situation du sujet, objectif du stage, approches expérimentales Les translocations chromosomiques, qu’elles soient réciproques ou Robertsoniennes, sont caractérisées par le fait que leur transmission dans les gamètes peut se faire de façon équilibrée, avec ou sans la translocation, ou déséquilibrée, entraînant ainsi, dans le produit de conception, des associations trisomies-monosomies partielles pour les chromosomes impliqués. Ces dernières aboutissent soit à des fausses couches soit à la naissance d’un enfant porteur d’un syndrome chromosomique grave. Chez l’homme, les proportions relatives des gamètes équilibrés et déséquilibrés peuvent être étudiées en FISH sur spermatozoïdes. Il a été montré une augmentation globale du taux de fragmentation de l’ADN spermatique chez les porteurs de translocations. Le but du travail proposé est d’utiliser une technique mise au point dans le laboratoire et permettant de visualiser en même temps le caractère fragmenté ou non de l’ADN d’un noyau de spermatozoïde et le type de ségrégation chromosomique chez des sujets porteurs d’une translocation chromosomique.
