IGEM- Chemotaxis July 15-21 Dose Dependant Swim Assay • Goal: – To measure the chemotactic response of bacteria to differing concentrations of toluene. • Procedure: – Plate a small number of bacteria onto center of a swim plates containing differing concentrations of toluene. – Measure the distance that the bacteria swim/grow in a 14 hour incubation period. Problems • Agar at the .15% concentration is EXTREMELY fluid. We could not invert plates for incubation or measurement/photgraphing. • Toluene is flammable and volatile. • Plating a consistent number of bacteria onto the swim plate is difficult with an inoculating loop Results Average Area (mm) pWW0 10mM 0.335 5mM 0.550 0mM 0.168 KT2440 10mM 5mM 0mM 0.507 0.528 0.104 Further Research • Use .3% agar (I calculated incorrectly) • Grow the colonies out in media, take an absorbance reading, and plate a small amount of a dilution to achieve a similar initial cell count among plates. • Larger # in order to verify results which do not agree with paper we used as guide. • Higher/Lower Concentrations • Gradiants
