Disinfection of all tissue culture equipment and work surfaces in the lab have to be performed regularly to prevent
contaminations with mycoplasmas and other unwanted
microorganisms, or to eliminate microbes once they have
been detected. PromoKine offers different disinfectants ideal
for laboratory surfaces and apparatuses including CO2 incubators and workbenches, as well as solutions for maintaining

BIOMYC-1 is based on the antibiotic tiamutin, which is
produced by the fungus Pleurotus mutilus. BIOMYC-2
is based on the antibiotic minocycline, an improved derivative of tetracycline. When used in combination these
two antibiotics eliminate mycoplasmas very efficiently.
Following our recommended protocol, the mycoplasmas
do not develop resistance to these antibiotics - a common
occurrence with other antibiotic treatment methods.

BIOMYC-3 is based on the antibiotic ciprofloxacin, which
inhibits the synthesis of DNA gyrase. Many mycoplasma
species are sensitive to BIOMYC-3, including M. orale, M.
hyorhinis, M. fermentans and M. arginini, and Acheloplasma laidlawii. These species are responsible for most
mycoplasma contaminations in cell culture.
At the concentrations recommended for use, no cytotoxic
effects have been found, and the treatment can be performed within 12 days.

The water in an incubator or in a water bath is a major
source of cell culture contamination, particularly with
respect to mycoplasmas. Aquaguard, a non-volatile water
stabilizer/cleaner, is extremely effective at maintaining the
water quality in CO2 incubators (tray) or ordinary water
baths. In addition, it does not cause damage to the stainless steel of the incubator or any plastic interior, and it does
not cause any skin irritation when used as recommended.

Detection of mycoplasmas utilizing a conventional light
microscope is nearly impossible. In addition, cloudiness of
media, typical for stronger bacterial or yeast contaminations, does not occur. PCR on the other hand is a highly sensitive, specific, and rapid method for the detection of even
very low mycoplasma or bacterial contamination levels in
culture supernatant, cell/virus stocks, or medium.

PCR Mycoplasma Tests Kits I & II
PCR Mycoplasma Test Kit I/RT
The PCR Mycoplasma Test Kit I/C utilizes conventional
PCR technology for the convenient, specific and highly
sensitive detection of up to 40 mycoplasma and acholeplasma species, including all species known to contaminate
cell cultures. The test can be performed within 3 hours and
includes In­ternal Control DNA (as negative control and for
verifying a successful PCR run) as well as Positive Control
DNA. All components come opti­mally premixed and lyophilized in PCR tubes. After recon­stitution with Reaction
Buffer, the sample is added to this master mix and the PCR
can be started. The PCR Mycoplasma Test Kit II also includes all components required for PCR premixed at optimized
concentrations - however, in a ready-to-use (liquid) master
mix. It also includes Positive Control DNA but in contrast
to the PCR Mycoplasma Test Kit I/C it does not contain an
In­ternal Control DNA.
The PCR Mycoplasma Test Kits I/RT has been developed
for convenient, highly sensitive, fast and reproducible RealTime PCR detection of mycoplasma contamination. The
kit is compatible with any Real-Time PCR instrument able
to detect the fluorescence dyes FAM™ and HEX™, and
includes Internal Control DNA (for verifying a successful
PCR run) as well as Positive Control DNA. All components come optimally premixed and lyophilized in PCR tubes.
After reconstitution with Reaction Buffer, the sample (and
– for kit variants A and B – Hot-Start Taq polymerase)
is added to this master mix and the PCR can be started.
DNA standards to perform a quantitative PCR analysis are
available separately.

Mycoplasma Elimination
In addition, PromoKine provides a highly sensitive and
time-saving PCR Bacteria Test Kit for detection of contaminating bacteria in cell culture media and virus stocks.
It is applicable for 45 bacteria species and detects as little
as 12 bacterial genomes (~52 fg) per sample volume. The
ready-to-use kit includes positive and internal controls and
a convenient protocol for easy sample preparation.
Mycoplasma contaminations seriously affect cell growth
and cell physiology leading to biased and unreliable experimental results. Standard antibiotics commonly used in cell
culture (e.g. Pen/Strep) do not affect mycoplasma growth.
Therefore, PromoKine offers two highly effective methods
to get rid of mycoplasma contamination: Mycoplasma-EX,
a combination of non-antibiotic and antibiotic reagents,
and BIOMYC, three highly effective antibiotics.

Mycoplasma-EX combines a non-antibiotic and a thoroughly adjusted antibiotic treatment. It eliminates mycoplasmas by directly killing them and not just by inhibiting
their growth, while showing extremely low toxicity for the
cells in culture. Mycoplasma-EX is effective with only one
treatment, destroying mycoplasmas fast and permanently.
It is applicable for most cell lines (e.g. Vero, BHK21, GBK,
ML, Hep2, 293, CRFK, H9, Molt4, MT-4, Jurkat) as well
as virus stocks (e.g. SHV-1, BHV-1, HSV-1, VSV, SFV, FCV,
