It will involve the phenotypic characterisation of several inducible cell lines followed by the use of cutting edge approaches such as ATACseq or Massspectrometry. Finally, collaborations with clinicians will be developed Proposition de sujet de thèse à l’appui d’une demande de contrat doctoral 2017-2087 (L’ensemble de cette fiche ne doit pas dépasser 1 page) Nom, prénom du directeur de l'unité de recherche : Baldacci Giuseppe Numéro de l'unité de recherche (et établissement de rattachement) : UMR7592 Nom, prénom du responsable de l'équipe d'accueil (EAD) : Ribes Vanessa Nom, prénom du directeur de thèse : Ribes Vanessa Titre du sujet de thèse proposé : (en anglais) Dynamics in genomic landscapes triggered by PAXFOXO1 and underlying Rhabdomyosarma plasticity Citer 5 mots clés : (key words) Rhabdomyosarcomes, Cellular transformations, Transcriptional regulation, Protein partners, PAX transcription factors Candidat pressenti : OUI Line Manceau NON The project aims to make significant contributions in understanding the molecular mechanisms by which PAX3FOXO1 and PAX7FOXO1 fusion transcription factors trigger trans-differentiation into tumourigenic cells and cause Rhabdomyosarma (RMS). For this, we will use several PAXFOXO1 inducible cell lines which will allow us to tune 3 parameters: the levels of expression of the fusion genes, the types and the differentiation stage of cells in which they will be expressed. We plan to analyse the impact of these 3 parameters onto the transcriptome and phenotypic cell states by combining RNAseq experiments with classical histological analyses. Then, we will test whether the propensity of PAXFOXO1 protein to trigger transdifferentiation relies on pioneer properties, by notably examining the opening state of chromatin in PAXFOXO1 bound domains. Finally, we will fish out and determine the role of their protein partners in the control of the transcriptional state of cells; these proteins represent new drug targets to overcome the shortened life span of children affected by RMS.
