BIOL 201 - Assignment 4 - Due April 3, 2008 How differences in animal cell division are regulated EK Kieserman et al. (2008) Developmental regulation of central spindle assembly and cytokinesis during vertebrate embryogenesis. Current Biology 18:116-123 You may do this assignment by yourself or in groups of two or three. You may discuss this assignment with anyone you want but when writing this assignment only people listed as authors may be present. Your answers must be in your own words and not paraphrased from the article or from other sources. When writing your answers consider myself to be your audience. Assume that I am familiar with cell biology techniques and information but that I read this paper a year ago and have forgotten the details. Cite any sources used other than the article itself. Your assignment is to be single sided, use a cover page, and not be enclosed within a folder. You are encouraged to use hand drawn diagrams rather than computer drawn ones. This assignment is out of ****** marks and is worth 7.5% of your total course mark. Question 1 Figure 2D shows a cell in anaphase. Draw what the figure would have looked like if the authors had used antibodies against DNA and (i) Actin, (ii) Myosin, (iii) intermediate filaments, (iv) Condensin, and (v) Pom121. Use one diagram for each. (5 marks) Question 2 In a short paragraph describe the experiments shown in Figures 1A, 1B, and 1C and movies S1 and S2. Your answer must be in your own words and not those of the authors. Include in your answer the purpose of this experiment, how was it done, the results, and the conclusion. (5 marks) Question 3 Figures 2A-B and C-J look similar but there are two major differences in how these images were obtained. What are they? (2 marks) Question 4 In a short paragraph describe the experiment shown in Figures 3D and 3E and movies S9 and S10. Your answer must be in your own words and not those of the authors. Include in your answer the purpose of this experiment, how was it done, the results, and the conclusion. (5 marks) Question 5 Design an experiment to determine whether the amount of PRC1 has an affect on yeast mitosis. (Assume that yeast mitosis is regulated by PRC1 as well.) Your experiment should use a fluorescent microscopy base approach. You have access to any protein, cell type, reagent, or laboratory equipment you want. Include controls in your experiments and how you would interpret the results. Include a labeled diagram of possible results with your answer.
