Academic Scientists at Work Antisera Database System The Antisera Database File Storage system is designed to help you organize and find your reagents over the course of your lifetime. It is currently written as a FlieMaker Pro version 5.0 document for the Macintosh, but should be easily converted to a PC version. The file was designed in Dr. Boss’ laboratory and is in use. Modifications have been made to make it more general. The program can be modified and expanded, which you may wish to do. Importantly, the file can be used as it is. Below is a discussion of different Views and the fields: To enter a new antiserum into the database, click on the “New Record” button. Data Entry View 1 and 2 - There are two data entry views provided. All record fields are shared such that information typed into one record is in the other. Name of Antisera: Here you should assign a name that you will be able to place with a protein. Creator of page: The person who entered the data. Company, Catalog number, and Lot Number: These parameters are important if you will be reordering the same antisera. Lot number is VERY important in case a different lot does not provide you with the same data. Date Tested, Location of Western (or other assay) and the recommended standard dilution from that assay should be listed here. This will save you lots of time if this data is efficiently recorded. Because some antisera are homemade, other important pieces of information should be documented. Including Immunogen description and location: enter what was used to immunize the animal. Often homemade antibodies are purified to enrich the signal or reduce background. Some common methods are listed. E. coli cleared indicates that the antiserum was passed over a column bound with bacterial proteins. Ig purified indicates that the antibodies were purified with a protein A or protein G resin. Affinity purified indicates that the antibodies were purified over a peptide or protein column. Date and Notebook page for these experiments should be listed so that you can go back a year or two later and refresh your memory about the quality of the antisera. The Sort and Find buttons will help you move through the database. List View – Allows you to print out all your oligos with their location. You can sort or use the find buttons to limit the set.
