The importance of microbial xylanases has increased in present scenario due to its immense biotechnological applications in mainly food, animal feed, paper and pulp industries. Efforts on cost reduction have directed towards increasing enzyme production by developing better microbial strains, efficient fermentation techniques and recovery systems. The purpose of this study was to collect and isolate filamentous fungi from soil and humus of different regions in Mato Grosso do Sul, and evaluate the production of xylanase. The results revealed that among the 26 isolated tested, 16 (61,5%) could produce clear zones as a result of xylan utilization after 48h of incubation. Among these 16 isolated, in SR medium the enzymatic activity was the highest for Aspergillus niger (9.38U/mg protein) and Aspergillus terreus (8.04U/mg protein); while Aspergillus flavus (12.43U/mg protein) and Emericella sp. (6.62U/mg protein) showed increase of enzymatic activity in Khanna medium. M5 and M8 medium didn’t induce xylanase production. This study contributes to catalogue soil fungi isolated in the state of Mato Grosso do Sul, and provides additional information to support future research about the industrial potential of these microorganisms.
