TITLE: ELODEA CELLS 05 Text: page 34-42 Purpose: To identify basic structures of plant cells. Materials: microscope ruler light water slide dropping bottle coverslip methylene blue stain paper towels paper pencil CAUTION!!!! Methylene blue stain is a very dark blue stain. It will probably NOT wash out of your clothing. It is not harmful to your skin but it will leave a stain. Wash off your skin with soap and water as soon as possible. Any remaining stain on your skin will eventually wear off. 1. WEAR A LAB APRON. 2. AVOID GETTING STAIN ON CLOTHING OR SKIN. 3. KEEP THE BOTTLE OF STAIN AWAY FROM THE EDGE OF THE TABLE. 4. IF A SPILL DOES OCCUR, NOTIFY YOUR TEACHER IMMEDIATELY. Procedure: 1. Put the clean slide on the lab table. Put a drop of water in the center of the slide. 2. Put one FRESH GREEN leaf from the elodea plant in the water on the 3. Apply the coverslip. 1. slide. Put the slide on the stage of the microscope and focus on the leaf at 40 X. SCAN THE SLIDE. Notice the arrangement of the cells. 5. Increase magnification to 100 X and focus in on a small area of the leaf. 6. Now DRAW a group of cells at 100 X. IDENTIFY and LABEL the following parts: A. cell wall - rigid outside covering on plant cells. B. cell membrane - thin layer inside the cell wall. C. cytoplasm - filling material inside cell. D. chloroplasts - TINY, round green structures containing chlorophyll. Chloroplasts float in the cytoplasm 7. Remove the slide from the microscope. Remove the coverslip. 8. Apply a small drop of stain to the leaf and let it soak in for about two minutes, then blot off the stain with a paper towel. 9. Add a drop of water to the stained leaf and apply a coverslip. 10. Examine the slide again at low and then high power. TRY to locate a cell with a nucleus and ADD this to your drawing. LABEL. A. nucleus - dark blue round structure inside the cell. 11. Remove the slide from the microscope and clean it. Put all materials away!!!!
